摘要
采用多肽固相合成法,以Rink Amide树脂为载体、Fmoc保护的氨基酸为原料、DIC/HOBt为缩合体系,按照生长抑素类似物TT-232的氨基酸序列从C端向N端顺次偶联,肽树脂裂解得到还原型TT-232粗品,环化后再经制备型RP-HPLC纯化,得到纯度为98%的TT-232。
Using a solid-phase polypeptide synthesis method with Rink Amide resin as a solid support,Fmoc -amino acids as raw materials and DIC/HOBt as a condensation system,the crude reduced somatostatin analogue TT-232 was synthesized according to its amine acid sequence from C-terminal to N-terminal by sequential cou- pling and cleavage of peptide resin. After cyclization,the crude somatostatin analogue TT-232 was purified by RP-HPLC with a final purity of 98%.
