金骨莲胶囊对兔膝关节骨关节炎保护作用的实验研究

Preventive Effect of Jingulian Capsule on Knee Osteoarthritis in Rabbits

目的:探讨金骨莲胶囊对家兔膝骨关节炎的保护作用。方法:30只家兔完全随机法分为A组、B组、C组三组,每组10只,按改良Hulth法全部家兔切除右膝关节前交叉韧带和右膝内侧半月板,A组术后第1天开始灌服生理盐水,7.5mL/kg,1次/d,连续灌服8周。B组术后第1天开始灌服金骨莲胶囊,150mg/kg,1次/d,连续灌服8周。C组术后第1天开始灌服盐酸氨基葡萄糖片剂150mg/kg,1次/d,连续灌服8周。8周后处死全部家兔,取右侧膝关节软骨行病理学苏木精-伊红、番红染色,依据染色切片进行Mankin评分评估膝关节软骨关节炎程度。RT-PCR法分析测定膝关节软骨中IL-1β基因表达水平。结果:B,C两组家兔股骨髁大体标本、软骨苏木精-伊红及番红染色与A组存在差异。Mankin评分及IL-1β基因相对表达量,B组和C组两组均低于A组(P<0.05),差异有统计学意义;Mankin评分及IL-1β基因相对表达量B组与C组之间差异无统计学意义(P>0.05)。结论:金骨莲胶囊对兔膝关节炎有一定的保护作用且与氨基葡萄糖对软骨的保护作用近似,降低IL-1β表达是金骨莲胶囊产生软骨保护作用的途径之一。

Objective：To investigate the preventive effect of Jingulian capsule on knee osteoarthritis in rabbits. Methods： Thirty rabbits were randomly divided into group A,group B,and group C （n=10 in each group）. All of the rabbits underwent the excision of anterior cruciateligment and the medial meniscus on the right knee joint. Jingulian capsule （150 mg/ kg,once per day, for 8 weeks） were drenched to rabbits in group B at the first day after the operation, meanwhile normal saline （7.5 mL/kg, once per day, for 8 weeks） gave to rabbits in group A orally. Glucosamine hydrochloride tablets （150 mg/kg,once per day,for 8 weeks） were drenched to rabbits in group C at the first day after the operation. All rabbits were killed by gas embolism at the eighth week. Macroscopic evaluation was performed on the articular cartilage of medial malleolus of femur. Real Time-PCR （RT-PCR） was performed to investigate the expression of IL-1β in articular cartilage. Histologic evaluation was done according to the Mankin grading scale. Results： Expressions of IL-1β and mankin＇s scores were higher in group A than the other two group （P〈0.05）. There is no difference between group B and group C for expressions of IL-1β and mankin＇s scores（P〉0.05）. Conclusion：Jingulian capsule has preventive effect which is similar to glucosamine hvdrochloride tablets on knee osteoarthritis in rabbits.
