苦参碱注射液对抗FcεRI抗体阳性CAU病人血清致肥大细胞脱颗粒作用 被引量:5

摘要 目的探讨苦参碱注射液对抗高亲和力IgE受体(FcεRI)抗体阳性慢性自身免疫性荨麻疹(CAU)病人血清致肥大细胞(HMC)脱颗粒的影响。方法将密度为1×105/L的肥大细胞悬液100μL加入96孔细胞培养板,其中30孔每孔加入苦参碱注射液50μL(含苦参碱500μg)孵育24h为实验A组,不加苦参碱注射液30孔为实验B组,同时设加入苦参碱注射液50μL(含苦参碱500μg)30孔作为对照组。取10例抗FcεRI抗体阳性的CAU病人血清50μL,分别加入实验A组与B组孔中,10例健康人血清加入对照组孔中。孵育20min后,观察各组组胺释放量及肥大细胞脱颗粒率。结果实验A组、对照组组胺释放量、肥大细胞脱颗粒百分率均低于实验B组,差异有显著性(F=165.081、43 688.714,P<0.05)。结论苦参碱注射液在体外对含抗FcεRI抗体阳性的CAU病人血清致肥大细胞脱颗粒具有抑制作用。 Objective To explore the impact of matrine injection on mast cell degranulation caused by serum of patients with chronic autoimmune urticaria(CAU)with positive anti-FcεRI body. Methods Mast cell suspension with a density of 1×105/L-100μL-was added in a 96-hole cell culture plate,of which,50μL of matrine injection(containing matrine 500μg)were added in each hole and incubation for 24h(group A),the other 30 holes added in no-matrine injection(group B),and the other 30 holes were added in 50μL(containing matrine 500μg)served as control group.Serum of the 10 CAU patients was collected and added 50μL in groups A and B,respectively,and serum of 10 healthy subjects was added in the control group.After 20 minutes of incubation,histamine release(HR)and mast cell degranulation rate(MCDR)were observed. Results In group A,MCDR and HR were lower than that in group B,the difference was significant(F=165.081,43 688.714;P〈0.05). Conclusion Matrine injection can inhibit the mast cell degranulation,in vitro,caused by serum of chronic autoimmune urticaria patients with positive anti-FcεRI body.
