牛磺酸对氟致神经细胞内钙离子浓度和细胞凋亡改变的干预研究 被引量:4

The intervention of taurine on the changes of fluorescent intensity of Ca^(2+) and apoptosis in neuron induced by sodium fluoride

摘要 目的通过观察牛磺酸对氟化钠(Na F)致神经细胞内钙离子(Ca^(2+))浓度和细胞凋亡改变的干预作用,为探讨氟中毒的机制及其预防提供实验依据。方法出生7 d昆明种小鼠乳鼠腹腔注射不同剂量的牛磺酸溶液,于注射后2 h在腹腔注射氟溶液。实验共分4组,即空白对照(生理盐水);氟组(Na F 2.8 mg/kg·bw);低剂量牛磺酸加氟组(牛磺酸7.5 mg/kg·bw+Na F 2.8 mg/kg·bw);高剂量牛磺酸加氟组(牛磺酸15.0 mg/kg·bw+Na F 2.8 mg/kg·bw)。激光扫描共聚焦显微镜记录细胞内Ca^(2+)荧光图像,流式细胞术检测细胞凋亡。结果牛磺酸可以降低Na F引起的神经细胞内Ca^(2+)的升高,高剂量牛磺酸组Ca^(2+)荧光值明显降低,与对照组比较差异有统计学意义(P<0.01)且牛磺酸组与氟组之间差异亦有统计学意义(P<0.01);同时牛磺酸还可明显抑制由于Na F影响所导致的神经细胞凋亡,与对照组比较,差异有统计学意义(P<0.01)且牛磺酸组与氟组比较亦有统计学意义(P<0.01)。结论在本实验条件下,适当剂量的牛磺酸可在一定程度上改善氟所致神经细胞钙超载及神经细胞凋亡等损害。 Objective To explore the intervention of taurine on the changes of fluorescent intensity of Ca^(2+)and apoptosis in neuron induced by sodium fluoride,and to provide the experimental basis on mechanic research and prevention for fluoride-induced toxicity.Methods New born mice( 7d) were injected intraperitoneally with different doses of taurine and then fluoride after 2 h. The experiment was divided into 4 groups,control group( normal saline),Na F group( Na F 2. 8 mg / kg·bw),high taurine plus Na F group( taurine 7. 5 mg /kg·bw + Na F 2. 8 mg / kg·bw) and low taurine plus Na F group( taurine 15. 0 mg / kg·bw + Na F 2. 8 mg / kg·bw). Fluorescene of Ca^(2+)in cell were inspected by confocal laser scanning microscopy and apoptosis was detected by flow cytometry. Results Taurine could decreased the increased Ca^(2+)induced by Na F and fluorescence. There were significant differences( P 0. 01) compared with the control and Na F groups.Meanwhile,taurine could inhibited the neural apoptosis induced by Na F. There also were significant differences( P 0. 01) compared with the control and Na F groups. Conclusion Taurine could interfere neurotoxicity damage induced by fluoride.
