不同类型试剂盒提取血液基因组DNA效果的比较 被引量:4

Comparison of extraction effects of blood DNA by various kinds of kits

摘要 目的比较溶液沉淀法、离心柱法和磁珠法试剂盒提取血液基因组DNA的效果。方法采集高血压患者的血液样本90份,随机分为3组,分别采用溶液沉淀法、离心柱法和磁珠法试剂盒提取血液样本基因组DNA。通过紫外分光光度计、琼脂糖凝胶电泳检测及PCR扩增法对血液样本基因组DNA的提取效果进行比较。结果溶液沉淀法提取的DNA浓度显著高于离心柱法和磁珠法(P<0.05),离心柱法与磁珠法提取的DNA浓度差异无统计学意义(P>0.05),3种方法提取血液样本基因组DNA的A_(260)/A_(280)值及A_(260)/A_(230)值的差异均无统计学意义(P>0.05);3种方法提取的血液样本基因组DNA的大小基本相同,离心柱法提取的DNA点样孔处较干净,电泳条带单一,无拖尾;3种方法提取血液样本基因组DNA的PCR产物均可见单一且明亮的目的条带。结论 3种类型提取试剂盒各有优缺点,使用时应根据实际情况进行选择。 Objective To compare the effects of solution precipitation,centrifugal column and magnetic bead kits in extraction of blood genomic DNA.Methods Ninety blood samples from patients with hypertension were randomly divided into three groups,and determined for genomic DNA by solution precipitation,centrifugal column and magnetic bead kits respectively,of which the effects were compared by UV spectrophotometry,agarose gel electrophoresis and PCR amplification.Results The concentration of DNA extracted by solution precipitation kit was significantly higher than those by centrifugal column and by magnetic bead kits(P〈0.05).However,the concentrations of DNA extracted by centrifugal column and magnetic bead kits showed no significant difference(P〉0.05).The A_(260)/A_(280) and A_(260)/A_(230) values of DNA extracted by the three kits showed significant difference(P〉0.05).The lengths of DNAs extracted by the three kits were basically identical.However,the wells for loading of samples of DNA extracted by centrifugal column kit were clean,while single band without tailing was shown on electrophoretic profile.Each of PCR products of DNA extracted by the three kids showed single and bright target band.Conclusion The three kinds of DNA extraction kits showed individual advantages and disadvantages,which should be selected according to the actual situation.
