摘要
目的观察CYP2W1在胃癌组织中的表达,探讨其对胃癌细胞增殖和侵袭的影响。方法采用免疫组织化学技术,检测326例胃癌组织及正常胃黏膜组织中CYP2W1蛋白的表达;采用半定量反转录.聚合酶链反应(RT—PCR)实验检测随机选取10例胃癌组织和相应的正常胃黏膜组织中CYP2W1 mRNA的表达;构建4组胃癌及正常胃黏膜细胞株,应用Westernblot方法检测各组细胞株中CYP2WI蛋白的表达;使用平板克隆、划痕愈合实验研究其对胃癌细胞增殖和迁徙侵袭能力的影响。结果胃癌组织中CYP2W1蛋白(26.7%比0.0%)明显高于正常胃黏膜组织(X2=100.396,P〈0.05)。胃癌组织中CYP2W1mRNA(0.413±0.026比0.074±0.005)明显高于正常胃黏膜组织(t=28.115,P〈0.05)。胃癌细胞中CYP2W1蛋白的表达(0.481±0.024比0.000)明显高于正常胃黏膜细胞(t=49.097,P〈0.05)。CYP2W1阳性胃癌细胞的克隆形成的数量明显高于CYP2W1阴性细胞(P〈0.05)。24h/48hCYP2WI阳性胃癌细胞划痕修复速率明显高于CYP2W1阴性胃癌细胞,CYP2W1阳性胃癌细胞划痕愈合面积明显高于CYP2W1阴性胃癌细胞(P〈0.05)。结论CYP2W1仅在胃癌组织中表达,在正常胃黏膜组织中不表达;CYP2W1与胃癌细胞的生长增殖及迁徙侵袭能力明显相关。
Objective To investigate the expression of cytochrome P450 2W1 ( CYP2W1 ) in gastric cancer and its effect on proliferation and invasion. Methods The expressions of CYP2WI protein in 326 adenocarcinoma of gastric structures and in the corresponding 326 normal gastric mucosa structures will be tested by applying immunohistochemical technique. The expressions of CYP2W1 mRNA in 10 random chosen examples of adenocarcinoma of gastric structures and its corresponding normal gastric mucosa structures will be tested by semi -quantitative reverse transcriptase -polymerase chain reaction (RT -PCR). 4 groups of adenocarcinoma of gastric structure and normal gastric mucosa cell line will be constructed with the technology of Western blotting. The expression quantity of CYP2W1 in every group of cell line will be tested and its correlation will be analyzed. The influence of CYP2W1 expression towards the proliferation and invasion capacity of gastric cancer cell will be researched with the flat cloning experiment and scratch healing test. Results CYP2W1 protein ( 26. 7% vs. 0. 0% ) in the gastric cancer structures clearly is more than that in normal gastric mucosa structure(x2 = 100. 396, P 〈0. 05). CYP2W1 mRNA (0. 413 ± 0. 026 vs. 0. 074 ± 0. 005 ) in the gastric cancer structures clearly is more than that in normal gastric mucosa structure( t = 28.115, P 〈0. 05 ). The result of Western blotting shows that the expression quantity of CYP2W1 protein in the gastric cancer cell lines (0. 481 ±0. 024 vs. 0. 000) is obviously higher than that in normal gastric mucosa cell lines ( t = 49. 097, P 〈 0. 05 ). The result of flat cloning formation shows that the cloning - making quantities of gastric cancer cell in the experimental group are clearly higher than those in control group ( P 〈 0. 05 ). The result of scratch healing experiment shows that the scratch repair rate of gastric cancer cell after scratching for 24 hours/48 hours in experimental group is much higher than that in control group. The scratch healing proportion of gastric cancer cell in experimental group is much larger than that in control group ( P 〈 0. 05 ). Conclusion CYP2WI only expresses in the gastric cancer structure. However, it doesn' t express in the normal gastric mucosa structure; CYP2W1 is closely tied up with the growth multiplication capacity and motive invasion and metastasis capacity.
