摘要
[目的]筛选出适合金线莲各个组培阶段的组培快繁配方。[方法]试验以福建金线莲茎段为外植体,进行组织培养快繁技术及移栽技术研究。采用对比试验研究不同激素浓度的配比对金线莲茎段诱导、丛生芽增殖、生根壮苗的影响,并研究了移栽基质对金线莲移栽的影响。[结果]研究结果表明,MS+NAA 0.5 mg/L+BA 2 mg/L+蔗糖20 g/L+琼脂6 g/L最适合金线莲茎段诱导;MS+NAA 0.5 mg/L+BA 2 mg/L+KT 1 mg/L+蔗糖25 g/L+琼脂6 g/L最适合金线莲丛生芽增殖;MS+IBA 1.0 mg/L+NAA 1.0 mg/L+活性炭1 g/L+香蕉泥50 g/L+蔗糖25 g/L+琼脂6 g/L最适合金线莲壮苗生根;以泥炭土∶细沙=3∶1为栽培基质,其成活率最高。[结论]研究结果为金线莲工厂化生产提供了借鉴。
[Objective] The aim was to screen out rapid propagation formula for Anoectochilus roxburghii (Wall)Lind in various tissue culture p|mses. [Methodl This experiment used the stem as explants for studying the tissue culture for rapid propagation and transplanting techniques in the A noectochius roxburghii(Wall) Lind.. By comparison experiments, the effects of the hormone concentration ratios on the stem section induced, and multiple shoot clumps proliferation and rooting were analyzed, and the transplanting matrix for the transplanting were also compared. [Resultl The research results showed that: MS+NAA 0.5 mg/L+BA 2 mg/L+ sucrose 20 g/L +agar 6 g/L was best for the stem section induced. MS+NAA 0.5 mg/L+BA 2 mg/L+KT 1 rag/L+ sucrose 25 g/L+agar 6 g/L was best for multiple shoot clumps proliferation; 1MS+IBA 1.0 mg/L +NAA 1.0 mg/L+AC 1 g/L+ banana mud 50% + sucrose 25 g/L + agar 6 g/L was best for rooting;the cultivation substrate of peat soil :sand =3:1 was the most suitable for the highest survival rate.
