系统性红斑狼疮患者树突状细胞CD200R下调意义 被引量:1

Significance of CD200R Down Regulation on Dendritic Cells in Systemic Lupus Erythematosus

摘要 目的探讨系统性红斑狼疮(systemic lupus erythematosus,SLE)患者树突状细胞(dendritic cell,DC)表面CD200R表达及脂多糖(lipopolysaccharide,LPS)刺激单核细胞来源树突状细胞(monocytes derived dendritic cells,Mo DC)后其功能及CD200R表达的改变。方法收集SLE患者及健康对照各22例,利用流式细胞术、酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)及同种异体混合淋巴反应(mixed lymphocytes reaction,MLR)检测外周血DC表面CD200R的表达、LPS刺激对Mo DC活化及CD200R表达的影响。结果 SLE患者髓样树突状细胞(myeloid dendritic cell,m DC)及浆样树突状细胞(plasmacytoid dendritic cell,p DC)CD200R表达均低于健康对照(P<0.05),且m DC的CD200R表达与红细胞沉降率呈负相关(r=-0.552,P=0.041)。Mo DC经LPS刺激活化后CD200R表达下调(P<0.05),活化成熟标记HLA-DR、CD83、CD86表达上调(P<0.05),白细胞介素(interleukin,IL)-6和IL-10分泌增加(P<0.05),促CD4+T细胞增殖增加(P<0.05),SLE和健康对照未见显著性差异。结论 SLE患者DC表面CD200R表达下调,可能与其疾病发生及炎症状态关系密切。 Objective To analyze the expression of CD200 R on different subsets of dendritic cells in systemic lupus erythematosus( SLE) patients,and to investigate CD200 R changes during lipopolysaccharide( LPS)inducing monocytes derived dendritic cells( Mo DC) activation. Methods Peripheral blood samples were collected from 22 patients with SLE and 22 age and sex matched healthy volunteers. CD200 R expression on dendritic cells was determined by flow cytometry. Mo DCs were cultured and stimulated by LPS,then-the change of CD200 R expression and Mo DCs' activation were analyzed by flow cytometry,ELISA and mixed lymphocytes reaction( MLR). Results CD200 R was expressed in low level on plasmacytoid dendritic cell( p DC) and myeloid dendritic cells( mD C) in SLE( P 〈0. 05),CD200 R expression on mD C of SLE was negatively correlated to ESR. LPS could stimulate Mo DCs maturation and induce CD200 R down regulation both in healthy controls and SLE( P 〈0. 05). Conclusion CD200 R is down regulated in dendritic cells in SLE and might be related to the disease pathogenesis and inflammation.
