中药多糖对免疫抑制小鼠T细胞亚群影响的差异研究 被引量:12

摘要 目的比较巴戟天多糖(BJT)、酒黄精多糖(JHJ)、淫羊藿多糖(YYH)、人参多糖(RS)、黄芪多糖(HQ)、何首乌多糖(HSW)、生地多糖(SD)、枸杞多糖(GQ)对免疫抑制小鼠T细胞亚型影响的差异。方法采用环磷酰胺腹腔注射制成免疫抑制小鼠模型,通过流式细胞技术(flow cytometry,FCM)检测T细胞比例及分型。结果给予环磷酰胺后,与正常组比小鼠外周血中CD3+T细胞比值明显升高(P<0.01),YYH,高剂量HQ,低、高剂量RS、HSW、SD、GQ多糖对CD3+T细胞比值具有恢复作用(P<0.05);多糖对环磷酰胺所致Th1/Th2平衡失调具有恢复作用,其中YYH低剂量组,RS、HQ、JHJ、GQ低、高剂量组,HSW、SD高剂量组与模型组比较,差异具有统计学意义(P<0.05);而JHJ、GQ低、高剂量,HSW、SD高剂量多糖对Th2的影响较明显;此外,低、高剂量BJT、YYH、RS、GQ多糖对环磷酰胺所致Treg比例减少有恢复作用,与模型相比差异具有统计学意义(P<0.05或P<0.01)。结论补益类中草药多糖具有调节免疫系统平衡的作用,在这一过程中,性味不同中药来源的多糖可能通过作用于不同的免疫细胞起作用,滋补阴血中药的多糖免疫调节活性主要偏向于Th2细胞,温补阳气中药的多糖对Treg细胞作用较明显。 Objective To compare the differences of morinda polysaccharide (BJT),wine sibiricum(JHJ), epimedium (YYH), ginseng polysaccharides (RS), astragalus polysaccharide (HQ), polygonum polysaccharide (HSW), habitat polysaccharide (SD),and lycium barbarum polysaccharide (GQ) in affecting on T cell subset in immunosuppressed mouse. Methods Immunosuppressed mice model was made by intraperitoneal injection of cyclophosphamide,the ratio and subtype of T cell were detected by flow cytometry (FCM). Results After administration of cyclophosphamide,the ratio of CD3 cells in peripheral blood in mice was significantly higher(P〈 0.01);compared with the control group,low dose of YYH,high dose of HQ,low and high dose polysaccharide of RS,HSW,SD,GQ had a recovery ratio effect on CD3 cell (P〈0.05);polysaccharides had recovery ratio effect on Thl/Th2 imbalance induced by cyclophosphamide,which the difference was statistically significant in low and high dose polysaccharide of YYH,RS,HQ,JHJ,GQ,high dose polysaccharide of HSW,SD compared with "the model (P〈0.05);however,low and high dose polysaccharide of JHJ,GQ,high dose polysaccharide of HSW,SD had an obvious effect on Th2;in addition,low and high dose polysaccharide of BJT,YYH,RS,GQ had a recovery ratio effect on Treg induced by cyclophosphamide,compared with the model group,the difference was statistically significant (P〈0.05,or P〈0.01). Conclusion Tonic Chinese herb polysaccharides have effect of regulating the balance of immune system,in this process,the function of polysaccharide derived from Chinese medicine with different properties and taste reflected in effecting on difference immune cell,and the immunoregulatory activity of polysaccharides derived from Chinese medicine with nourishing Yin and blood is mainly related to Th2 cells,in addition,the polysaccharides derived from Chinese medicine with warming Yang-qi have obvious effect on Treg cell.
