摘要
目的 :探讨发状分裂相关增强子1(hairy and enhancer of split 1,Hes1)基因表达对食管腺癌OE33细胞增殖、侵袭及迁移的影响。方法 :应用实时荧光定量PCR及蛋白质印迹法检测人食管黏膜上皮HEEC细胞、食管鳞癌TE-1和食管腺癌OE33细胞中Hes1基因的表达水平。将特异性针对Hes1基因的sh RNA重组慢病毒感染食管腺癌OE33细胞,然后应用实时荧光定量PCR及蛋白质印迹法检测Hes1基因的沉默效率,并通过CCK-8、划痕愈合实验以及Transwell迁移和侵袭实验分别检测Hes1基因沉默对OE33细胞增殖、迁移及侵袭能力的影响。结果 :与HEEC和TE-1细胞相比,OE33细胞中Hes1 m RNA和蛋白的表达水平明显较高(P值均<0.01)。Hes1-sh RNA重组慢病毒感染OE33细胞后,Hes1 m RNA和蛋白表达水平均明显下调(P值均<0.01),并且细胞的增殖、迁移及侵袭能力均明显降低(P值均<0.01)。结论 :沉默Hes1基因的表达能一定程度上抑制OE33细胞的增殖、迁移及侵袭,提示Hes1可能是一个潜在的食管腺癌治疗靶点。
Objective:To investigate the effects of hairy and enhancer of split 1(Hes1) gene expression on the proliferation,migration and invasion of esophageal adenocarcinoma OE33 cells.Methods:The expression levels of f/es1 gene in human esophageal epithelial HEEC cells,esophageal squamous cell carcinoma TE-1 cells and esophageal adenocarcinoma OE33 cells were detected by realtime fluorescent quantitative PCR and Western blotting,respectively.The recombinant lentivirus with specific shRNA targeting f/esl gene was infected into esophageal adenocarcinoma OE33 cells,then the silenced efficiency of f/esl gene was detected by real-time fluorescent quantitative PCR and Western blotting,respectively.The effects of Hes1 gene silencing on the proliferation,migration and invasion abilities of OE33 cells were detected by CCK-8 assay,wound healing assay and Transwell migration and invasion assay,respectively.Results:The expression levels of Hes1 mRNA and protein in OE33 cells were significantly higher than those in HEEC and TE-1 cells(all P〈0.01).After infection with Hesl-shRNA recombinant lentivirus,the expression levels of Hesl mRNA and protein in OE33 cells were significantly reduced(both P〈0.01).Furthermore,the proliferation,migration and invasion abilities of OE33 cells were significantly inhibited after Hes1 gene silencing(all P〈0.01).Conclusion:Silencing the expression of f/esl gene can inhibit the proliferation,migration and invasion of OE33 cells,which indicates that Hesl may be a potential therapeutic target for esophageal adenocarcinoma.
