摘要
目的探讨淫羊藿苷(icariin,ICA)对野百合碱(monocrotaline,MCT)诱导的肺动脉高压(pulmonary arterial hypertension,PAH)模型大鼠血流动力学的影响。方法将48只雄性SD大鼠随机分为对照组、模型组、ICA(20、40、80 mg/kg)组和西地那非(Sildenafil,Sil,25 mg/kg)组,每组8只。一次性皮下注射MCT(50 mg/kg)复制PAH模型,7 d后按分组灌胃给药,每天1次,连续21 d。导管法测定心率(heart rate,HR)、颈动脉收缩压(carotid artery systolic pressure,CASP)、舒张压(carotid artery diastolic pressure,CADP)和平均压(carotid artery mean pressure,CAMP),肺动脉收缩压(pulmonary artery systolic pressure,PASP)、舒张压(pulmonary artery diastolic pressure,PADP)和平均压(mean pulmonary artery pressure,MPAP),右心室收缩压(right ventricle systolic pressure,RVSP)、舒张压(right ventricle diastolic pressure,RVDP)和平均右心室压(mean right ventricle pressure,MRVP),右心室舒张末期压(right ventricular end-diastolic pressure,RVEDP),心动周期(cycle duration)、收缩持续时间(systolic duration)和舒张持续时间(diastolic duration)以及右心室压最大上升(RV-Max d P/dt)和下降速率(RV-Min d P/dt)。结果与对照组比较,模型组大鼠HR、CASP、CADP和MCAP未见明显变化,PASP、PADP、MPAP、RVSP、RVDP、MRVP、Max d P/dt和Min d P/dt绝对值均显著增高,心动周期虽无显著改变,但收缩期明显延长,舒张期缩短。给予ICA可降低模型动物的HR、PASP、PADP、MPAP、RVSP、RVDP、MRVP、RV-Max d P/dt和RV-Min d P/dt绝对值,缩短收缩期,延长舒张期。结论 ICA可缓解MCT所致的PAH模型大鼠肺循环血流动力学异常。
Objective To investigate the effects of icariin(ICA) on hemodynamics of pulmonary arterial hypertension(PAH) induced by monocrotaline(MCT) in rats.Methods Forty-eight male SD rats were randomly divided into the control group,model group,ICA(20,40,80 mg/kg) and sildenafil(Sil,25 mg/kg),with 8rats in each group.PAH model was induced by MCT(50 mg/kg) singly subcutaneous injection.On the 7th days after MCT-injection,the animals were administrated intragastrically with ICA according to the above group protocol,once a day,for 21 days.The hemodynamic parameter,including heart rate(HR),carotid artery systolic pressure(CASP),carotid artery diastolic pressure(CADP),carotid artery mean pressure(CAMP),pulmonary artery systolic pressure(PASP),pulmonary artery diastolic pressure(PADP),mean pulmonary artery pressure(MPAP),right ventricular systolic pressure(RVSP),right ventricular diastolic pressure(RVDP) and mean right ventricular pressure(MRVP),right ventricular end-diastolic pressure(RVEDP),right ventricle cycle duration,systolic duration and diastolic duration,RV-Max d P/dt and RV-Min d P/dt,were measured by catheters placed in carotid artery or right ventricle or pulmonary artery.Results Compared with the control group,HR,CASP,CADP and MCAP showed no obvious changes,PASP,PADP,MPAP,RVSP,RVDP,MRVP,RV-Max d P/dt and absolute value of RV-Min d P/dt were significantly increased,right ventricle systolic duration was markedly prolonged and diastolic duration was notably shortened in model group rats.HR,PASP,PADP,MPAP,RVSP,RVDP,MRVP,RV-Max d P/dt and absolute value of RV-Min d P/dt were decreased,right ventricle systolic duration was markedly shortened and diastolic duration was notably prolonged in ICA treatment groups.Conclusion ICA can alleviate the abnormal pulmonary circulation hemodynamics in PAH model rats induced by MCT.
