皮肤鳞状细胞癌、基底细胞癌与日光性角化病皮损中组蛋白乙酰化的检测 被引量:1

摘要 目的研究皮肤鳞状细胞癌、基底细胞癌与日光性角化病中组蛋白H3、H4乙酰化的修饰水平。方法经HE染色病理确诊,收集21例皮肤鳞状细胞癌、22例基底细胞癌、19例日光性角化病及18例正常皮肤组织,应用ELISA法检测组蛋白H3、H4乙酰化表达水平。结果 1各组间组蛋白H3的乙酰化表达水平比较具有统计学意义(F=4.222,P<0.05);进一步两两比较发现,3组疾病中组蛋白H3的乙酰化表达水平均低于正常对照组,其中皮肤鳞状细胞癌(0.150±0.062)和正常皮肤对照组(0.279±0.164)相比差异具有统计学意义(P<0.05),基底细胞癌(0.187±0.102)、日光性角化病(0.196±0.121)与正常皮肤对照组相比差异没有统计学意义(P均>0.05);3组疾病之间两两比较差异均没有统计学意义(P均>0.05);2各组间组蛋白H4的乙酰化表达水平比较具有统计学意义(F=80.354,P<0.05);进一步两两比较发现,3组疾病中组蛋白H4的乙酰化表达水平均低于正常皮肤对照组,并且与正常皮肤对照组(3.515±0.727)相比差异均有统计学意义(P均<0.05);与皮肤鳞状细胞癌(0.547±0.065)相比,基底细胞癌(1.242±0.133)、日光性角化病(1.858±0.458)中组蛋白H4的乙酰化表达水平均升高(P均<0.05),且日光性角化病高于基底细胞癌(P<0.05)。结论 1与正常皮肤对照组相比,组蛋白H3乙酰化在皮肤鳞状细胞癌、基底细胞癌及日光性角化病中的表达水平差异不具有统计学意义,可能H3乙酰化在皮肤鳞状细胞癌、基底细胞癌及日光性角化病发病机制中不占主导作用;2与正常皮肤对照组相比,皮肤鳞状细胞癌、基底细胞癌与日光性角化病中组蛋白H4乙酰化的表达水平明显下调,组蛋白低乙酰化状态可能是皮肤鳞状细胞癌、基底细胞癌及日光性角化病的发生、发展的因素之一。3组蛋白H4乙酰化在日光性角化病、基底细胞癌及皮肤鳞状细胞癌中的表达水平呈递减方式,提示肿瘤恶性程度越高,组蛋白乙酰化程度越低。 Objective To study the acetylation of histone H3 and H4 in the cutaneous squamous cell carcinoma(CSCC),basal cell carcinomas(BCC) and actinic keratosis(AKs).Methods The acetylation of histones H3 and H4 in 21 cases of CSCC,22 cases of BCC,19 cases of AKs and 18 normal controls were detected with ELISA method.Results The expression of histone H3 in tissues of CSCC,BCC,and AKs groups showed statistically significance(F = 4.222,P〈0.05),and the expression levels were lower than that of normal control groups.There is a statistically negative correlation between CSCC(0.150 ± 0.062) and normal groups(0.279 ±0.164) in the expression of histone H3(P〈0.05),however,there was no statistical significance in BCC(0.187 ± 0.102),AKs(0.196 ± 0.121) and normal groups in the expression of histone H3(P〈0.05);There is no statistical significance between CSCC,BCC and AKs groups in the positive rate of histone H3(P〈0.05).The expressions of histone H4 in tissues of CSCC,BCC,and AKs groups were significant(F = 80.354,P〈0.05),and the expression levels were lower than that of normal control groups.There is a statistically negative correlation of histone H4 in CSCC(0.547 ± 0.065),BCC(1.242 ± 0.133),AKs(1.858 ± 0.458) and normal groups(3.515 ± 0.727).Conclusion The expression of histone H3 between CSCC,BCC and AKs tissues shows no statistical significance,probably suggesting that the acetylation of histone H3 is not the leading cause in these diseases.The acetylation of histone H4 expression levels in CSCC,BCC and AKs tissue was significantly lower than normal skin tissue,suggesting histone H4 acetylation may be associated with the development of CSCC,BCC and AKs.The acetylation of histones H4 in CSCC,BCC and AKs tissue gradually decreased,it probably suggest that the higher malignant of the tumor the lower of the level of histone acetylation.
