摘要
目的研究白术挥发性成分,并对5种肿瘤细胞抑制活性进行了检测。方法用水蒸气蒸馏得到挥发油成分,运用GC-MS技术并结合配套数据库进行化学成分检索和鉴定;MTT法检测白术挥发油对SMMC7721和Hep G2肝癌细胞、A549肺癌细胞、MCF-7乳腺癌细胞及HT29结肠癌细胞的抑制率。结果共从白术挥发油中分离并鉴定了42种成分,其中含量最高的为苍术酮。白术挥发油对SMMC7721、Hep G2、A549、MCF-7和HT29肿瘤细胞的IC_(50)值分别为21.62μg·m L^(-1)、24.02μg·m L^(-1)、≥200μg·m L^(-1)、36.58μg·m L^(-1)、30.15μg·m L^(-1)。结论白术挥发油主要成分为苍术酮等倍半萜类化合物,其对五种癌细胞均有不同程度的抑制作用,其中对SMMC7721肝癌细丙抑制活性最强。
OBJECTIVE To study the chemical components of volatile oil from Atractylodes macrocephala Koidz and test their inhibitory activities on five tumor cells. METHODS The volatile oil grom Atractylodes macrocephala Koidz was extracted by steam distillation. Technology of GC-MS combined with a complete set of database was used to search and authenticate the volatile components; MTT method was used to test the inhibitory activity to the tumor cells of SMMC7721,Hep G2,A549,MCF-7 and HT29. Results 42 kinds of chemical composition were identified and the content of atractylone was the most. The IC_(50) of the volatile components to tumor cells of SMMC7721,Hep G2,A549,MCF-7 and HT29 were 21. 62μg ·m L^-1、24. 02μg·m L^-1、≥200μg·m L^-1、36. 58μg·m L^-1、30. 15μg·m L^-1respectively. CONCLUSION The main ingrdients of the volatile components of Atractylodes macrocephala Koidz were the sesquiterpenoids such as atractylone. The inhibitory activity to five tumor cells were different and the strongest inhibitory activity was SMMC7721.
