低浓度溶血磷脂酸诱导海马神经元凋亡及其对c-jun磷酸化的影响 被引量:1

摘要 目的分析低浓度溶血磷脂酸(LPA)诱导海马神经元凋亡的作用是否可被拮抗剂苏拉明阻断;探讨溶血磷脂酸对c-jun磷酸化的影响。方法 MAP2-DAPI双染免疫荧光鉴定海马神经元及测定纯度;流式细胞仪测海马神经元凋亡率:6孔板内培养第6天的海马神经元随机分为对照C组(加入等体积的去离子水)、L组(加入0.1μmol/L溶血磷脂酸)和L+S组(加入10μmol/L苏拉明+1μmol/L溶血磷脂酸);Western blot检测JNK、c-jun磷酸化水平及c-jun表达,6 cm培养皿里培养第6天的海马神经元随机分为L组(加入1μmol/L溶血磷脂酸)和正常对照C组(加入等体积的去离子水)。结果体外海马神经元培养成功,且海马神经元纯度检测为97%。L组凋亡率明显高于C组(P<0.05),而L+S组凋亡率与C组相比差异无统计学意义(P>0.05)。L组磷酸化JNK、磷酸化c-jun水平均较C组高,差异有统计学意义(P<0.05),c-jun的表达亦较C组显著增高(P<0.05)。结论 LPA通过与海马神经元表面的受体结合介导其诱导凋亡作用;凋亡浓度的LPA可以诱导JNK/c-jun激活,促进c-jun表达。
