摘要
目的:探讨急性高糖对缺血再灌注后大鼠肾组织髓过氧化物酶(MPO)和环氧合酶2(Cox-2)表达的影响。方法:成年雄性SD大鼠随机分为正常血糖假手术(NG-Sham)组、正常血糖缺血再灌注(NG-I/R)组、高糖假手术(HG-Sham)组、高糖缺血再灌注(HG-I/R)组4组(n=8)。大鼠切除右侧肾脏,夹闭对侧肾蒂45 min再灌注24 h制备肾缺血再灌注模型。大鼠急性高糖模型为术前45 min通过腹腔注射25%葡萄糖3 g·kg-1制备。比色法检测大鼠肾脏MPO,RT-PCR法检测肾脏Cox-2-mRNA,Western blotting检测肾脏Cox-2蛋白表达。结果:与Sham组相比,I/R组MPO活性,Cox-2-mRNA、Cox-2蛋白表达均显著增高(P<0.05),而HG-I/R组的MPO活性,Cox-2-mRNA、Cox-2蛋白表达又明显高于NG-I/R组(P<0.05)。结论:急性高糖可以增加缺血再灌注后大鼠肾组织MPO活性,增加Cox-2的表达水平,加重肾缺血及再灌注所引发的炎症损伤。
Objective: To investigate the effect of acute hyperglycemia on expression of MPO and Cox-2 in rats with renal ischemia reperfusion injury. Methods: Sprague-Dawley rats were randomly arranged to the normoglycemic( NG)- and hyperglycemic( HG) groups,with each group further divided into sham and I/R groups. Acute hyperglycemia was induced by intraperitoneal injection of 25% glucose( 3 g·kg^- 1) 45 min before ischemia. The right kidney of rats were removed,cliped the contralateral renal pedicle 45 min and reperfusion 24 h to establish the renal ischemia reperfusion model. The activity of MPO was measured with colorimetric method. Cox-2 were measured by RT-PCR and Western blotting. Results: Compared with group Sham,the activity of MPO,the expression of Cox-2-mRNA,Cox-2 protein were increased in group I / R( P〈0. 05),however,when compared with group NG-I /R,the activity of MPO,the expression of Cox-2-mRNA,Cox-2 protein were increased in group HG-I / R( P〈0. 05). Conclusions: The acute hyperglycemia increases MPO activity and the level of Cox-2 in kidney of ischemiareperfusion,which can aggravate the inflammatory injury caused by renal ischemia reperfusion injury.
