miR-101对结直肠癌细胞SW620生物学特性的影响 被引量:2

摘要 目的探讨miR-101对结直肠癌细胞SW620增殖、凋亡及细胞周期的影响。方法利用CCK8法、平板克隆形成实验、细胞周期、细胞凋亡方法分别检测SW620、GV209-SW620、mir101-SW620 3组细胞的生物学特性。结果 CCK8比色法结果显示SW620各组细胞生长时间水平差异具有显著性(F=4152,P<0.001),细胞增殖能力组间有显著性差异(F=10220,P<0.001)。平板克隆实验的结果为SW620空白对照组、GV209-SW620对照组和mir101-SW620组的克隆形成率分别为:44.33%、48.17%、35.17%。SW620 3组细胞间的克隆形成能力差异具有统计学意义(F=73.015,P<0.001)。细胞周期分析发现SW620各组细胞间其G1、S、G2/M期存在着统计学差异(F=45.974,P=0.019;F=122.139,P=0.000;F=115.171,P=0.000)。除了SW620和GV209-SW620的G2期分布无统计学差异外(P=0.441),其余各组G1、G2/M期均有统计学差异(P<0.01);与SW620和GV209-SW620相比,mir101-SW620组出现了明显的G1和G2/M期周期阻滞。细胞凋亡实验发现SW620细胞各组细胞凋亡率差异有统计学意义(F=6115,P<0.001),各组进行两两比较,均有统计学差异(P<0.01)。与SW620空白细胞组、GV209-SW620对照组相比,mi R-101过表达组细胞凋亡率增加(P<0.05)。结论 mi R-101能够抑制结直肠癌细胞SW620的增殖,细胞G1和G2/M期周期阻滞及促进细胞凋亡。 Objective To investigate the effect of mi R-101 on biological behaviors of colorectal cancer cell line SW620.Methods CCK-8 method,colony formation assay,cell cycle analysis and apoptosis analysis were applied to assess the effects of mi R-101 on cell proliferation,invasion and apoptosis of SW620 cells.Results Over-expression of mi R-101 in SW620 cells significantly suppressed the cell proliferation and attenuated the colony-forming ability of the cells.Flow cytometry showed that over-expression of mi R-101 in SW620 cells caused obvious cell cycle arrest in G2/M and G1 phases,and significantly increased the cell apoptosis rate.Conclusion Over-expression of mi R-101 can inhibit the proliferation,cause cell cycle arrest and promote apoptosis of colorectal cancer SW620 cells.
