GSH对MRL/lpr小鼠肾小管上皮细胞TGF-β1表达影响研究

摘要 目的:探讨还原性谷胱甘肽(GSH)对MRL/lpr小鼠肾小管上皮细胞内转化生长因子β1(TGF-β1)m RNA表达的影响,了解还原性谷胱甘肽对肾小管上皮细胞中转化生子因子β1表达的影响。方法:传代培养C57BL/6雌性小鼠及狼疮小鼠模型MRL/1pr雌性小鼠肾小管上皮细胞,给予第3代细胞还原型谷胱甘肽药物干预,培养5天后检测细胞内TGF-β1的表达。结果:狼疮小鼠模型MRL/1pr雌性小鼠肾小管上皮细胞内TGF-β1表达明显高于C57BL/6小鼠(P<0.05)。经还原性谷胱甘肽处理的MRL/1pr雌性小鼠肾小管上皮细胞表达TGF-β1明显低于未处理组(P<0.05)。结论:还原型谷胱甘肽干预可有效减少MRL/1pr雌性小鼠肾小管上皮细胞表达TGF-β1。 Objective: To investigate the effects of glutathione (GSH) on transforming growth factor betal (TGF- β1) mRNA expression in renal tubular epithelial cells of MRL/lpr mice. Methods: Renal tubular epithelial cells of G57BL/6 female mice and MRL/ lpr female mice were cultured and passaged to the third generation, respectively. Expression of TGF- β 1 mRNA in cells were evaluated after 5 days of culture with or without GSH. Results: TGF-β 1 mRNA expression in renal tubular epithelial cells of MRL/lpr female mice was significantly higher than that of G57BL/6 female mice (P 〈 0.05). Expression of TGF- β1 mRNA in renal tubular epithelial cells of MRL/Ipr female mice cultured with GSH was significantly lower than that without GSH (P 〈 0.05). Conclusions: GSH may result in decreasing TGF- β 1 expression in renal tubular epithelial cells of MRL/lpr female mice.
