摘要
目的:观察桑柏生发方对毛发再生过程中血管内皮生长因子(VEGF)等细胞因子的影响。方法:建立完善的体外培养毛乳头细胞技术体系,采用不同浓度的桑柏生发方提取(水提,醇提)液作用于体外培养的4-6代人毛乳头细胞72 h,用四甲基偶氮唑蓝(MTT)法测定体外培养的人毛乳头细胞增殖活性;ELISA法检测细胞上清液IL-1β、VEGF和TNF-α蛋白含量;反转录(RT)-PCR半定量检测细胞IL-1β、VEGF和TNF-αm RNA表达。结果:与空白对照组比较,桑柏生发方提取液均能显著促进人毛乳头细胞增殖(P<0.05),促进细胞VEGF的分泌与表达、抑制分泌IL-1β、促进分泌TNF-α(P<0.05)。结论:桑柏生发方提取液有刺激人毛乳头细胞增殖的作用,可能机制为促进细胞因子VEGF、TNF-α分泌,抑制IL-1β的分泌而刺激毛发再生。
Objective: To observe the influence of Sangbai Shengfa Formula on the germinal party vascular endothelial growth factor. Methods: We established a perfect system of cultivating dermal papilla cells in vitro, extracted different concentrations of the Sangbai Shengfa Formula on the vitro cultured dermal papilla cells 4th to 6th generations for 72 h, and measured the proliferation activity of vitro cultured human dermal papilla cells with MTT; The IL-1β, VEGF and TNF-ctprotein content in supernatant fluid were detected by ELISA method; the mRNA expression of IL-1β,VEGF and TNF-cdn cells were evaluated by Reverse transcription (RT) -PCR semi quantitative. Results: Compared with control group, Sangbai Shengfa Formula extract could significantly pro- mote the proliferation of human dermal papilla cells (P〈0.05), promote cell secretion and expression of VEGF, inhibit secretion of IL-1β, and promote the secretion of TNF-α (P〈0.05). Conclusion: Sangbai Shengfa Formula extract can significantly stimulate the role of human dermal papilla cells proliferation, the mechanism of which may be promoting by cytokine VEGF, inhibition of IL-1β, and promote the secretion of TNF-αto stimulate hair regeneration.
