摘要
目的:探讨毛蕊异黄酮在体外对人乳腺癌细胞MCF-7增殖和迁移的作用及其相关机制。方法:分别采用MTT法和Transwell小室测定毛蕊异黄酮对MCF-7细胞增殖和迁移能力的影响,对于细胞内VEGF蛋白水平的影响通过Western Blot进行检测。结果:毛蕊异黄酮呈剂量依赖性抑制了MCF-7细胞的增殖活性和迁移,与对照组相比,毛蕊异黄酮可明显降低MCF-7细胞中VEGF蛋白的表达(P<0.05)。结论:毛蕊异黄酮对MCF-7细胞的增殖和迁移有抑制作用,其机制可能与下调细胞中VEGF表达有关。
Objective: To investigate the effect of calycosin on the proliferation and migration of human breast cancer cell MCF-7 in vitro and its possible mechanism. Methods: Effect of calycosin on the proliferation of MCF-7 cells were determined by the MTT assay, while the migration by transwell assay. The protein level of VEGF in calycosin-treated cells was determined by Western Blot. Results: Compared with the control group, calycosin significantly inhibited the proliferation and migration of MCF-7 cells in a dose-dependent manner (P 〈 0.05). In addition, the protein level of VEGF in MCF-7 cells was down-regulated by calycosin (P 〈 0.05). Conclusion: Calycosin has an inhibitory effect on both proliferation and migration of MCF-7 cells, whose mechanism may be related to the down-regulated cells in the VEGF expression.
