摘要
为进一步探究绵羊肺炎支原体P108蛋白的结构及其免疫原性,在进行生物信息学分析的基础上,对编码P108的部分基因片段进行了PCR扩增、克隆和原核表达,并采用Western-blot方法对其免疫原性进行了分析。结果显示,p108基因与猪肺炎支原体黏附相关基因mhp385的核苷酸同源性较高,扩增的p108部分基因在大肠杆菌BL21(DE3)中成功获得表达,重组蛋白以包涵体的形式存在;重组蛋白可以与山羊抗绵羊肺炎支原体血清发生结合反应,表明P108蛋白是免疫原之一。本研究结果表明P108蛋白可以诱导机体产生免疫应答,是建立绵羊肺炎支原体感染血清学诊断技术的潜在抗原。
To investigate the structure and immunogenicity of P108 protein in Mycoplasma ovipneumoniae, based on bioinformatics analysis,a fragment of p108 gene was selected and expressed in Escherichia coli. The immunogenicity of P108 was analyzed by Western-blot. The results demonstrated that pl08 gene was a homologue to mhp385 in Mycoplasma hyopneumoniae. The part fragment of p108 gene was successfully expressed in L coli BL21(DE3) as the form of inclusion bodies. The recombinant protein could react with goat serum against M.ovipneuminae,indicating that P108 is one of the immunogens of M.ovipneumoniae, rhe results suggest that P108 can induce immune responses,therefore it might be used as a potential antigen for development of serological diagnostic tools for M.ovipneumoniae.
