摘要
为从分子水平上明确药用美洲大蠊在分类学上的地位,采用分子克隆和序列比对的方法,对云南大理产美洲大蠊的CO I基因和18S rRNA基因片段进行扩增、克隆和序列测定,并同蜚蠊科相关物种进行系统发育分析。结果表明,通过邻接法(Neighbor-joining method,NJ)和最小进化法(Minimum evolution method,ME)构建的系统树基本一致,证实CO I能对美洲大蠊及其近缘物种进行有效的物种分子鉴定,而18S r RNA不能进行有效的分子鉴定。
CO I and 18 S r RNA genes of Periplaneta americana collected at Dali,Yunnan Province were cloned and sequenced.The phylogenetic relationships between the Periplaneta americana and its closely related 5 species in the order Blattaria were analyzed.The results showed that the phylogenetic trees were almost identical with methods of NJ(Neighbor-joining) and ME(Minimum evolution).It showed that CO I gene was a practical molecular marker for species identification of Periplaneta americana and its closely related species,while 18 S r RNA gene failed to be used in species identification.
