盐炙对知母中新芒果苷体内药代动力学的影响 被引量:6

摘要 目的:建立测定大鼠血浆中新芒果苷HPLC方法,探究盐炙对知母中新芒果苷药代动力学的影响。方法:色谱柱:Ecosil C_(18)柱(250 mm×4.6 mm,5μm);流动相:水-乙腈(15:85);流速:1 m L/min;检测波长:258 nm;柱温:30℃。结果:新芒果苷在生品和盐炙品组大鼠体内药代动力学参数分别为:AUC_(0-t)为(43.913±0.636)、(71.565±1.950)mg/L*h;AUC_(0-∞)为(44.127±0.673)、(71.805±1.926)mg/L*h;C_(max)为(4.539±0.048)、(5.805±0.119)mg/L;MRT_(0-t)为(8.124±0.246)、(8.650±0.398)h;MRT_(0-∞)为(8.290±0.321)、(8.761±0.435)h;t_(1/2)为(4.315±0.472)、(3.989±0.301)h。结论:知母盐炙后促进新芒果苷的吸收,并在一定程度上增加了生物利用度。 Objective: To establish an HPLC method to determine neomangiferin in rat plasma and investigate the effect of salt processing on pharmacokinetics of neomangiferin in Anemarrhena Rhizoma. Methods: It was obtained with Ecosil column( 250 mm× 4. 6 mm,5 μm) and eluted with acetonitrile and water( 15: 85) and its flow rate was 1 m L/min. The detection wavelength was set at 258 nm and the column temprature 30 ℃. Results: The main pharmacokinetic parameters neomangiferin in raw and salt products: AUC_( 0- t)were( 43. 913 ± 0. 636)( mg/L* h) and 71. 565 ± 1. 950( mg/L* h); AUC_( 0- ∞)were( 44. 127 ± 0. 673)( mg/L* h) and 71. 805 ± 1. 926( mg/L* h); C_(max)were( 4. 539 ± 0. 048)( mg/L) and 5. 805 ± 0. 119( mg/L); MRT_( 0- t)were( 8. 124 ± 0. 246) h and( 8. 650 ± 0. 398) h; MRT_( 0- ∞)were( 8. 290 ± 0. 321) h and( 8. 761 ± 0. 435) h; t_(1/2)were( 4. 315 ±0. 472) h and( 3. 989 ± 0. 301) h. Conclusion: Salt processing can obviously enhance the absorption of neomangiferin in Anemarrhena Rhizoma and increase the bioavailability.
