青蒿琥酯对皮肤癌细胞核形态变化及AgNOR颗粒形态的影响

摘要 目的:研究青蒿琥酯对皮肤癌细胞核形态变化影响及Ag NOR颗粒形态影响。方法:购买人表皮鳞癌细胞株A431细胞株,随机分为两组,常规培养后,200μmol/L的青蒿琥酯溶液(经10%胎牛血清稀释)1 m L;对照组加入等量的10%胎牛血清,培养48 h后制作切片;采用HE染色和银染色处理切片。通过彩色病理图像分析系统观察两组细胞核形态、细胞结构、Ag NOR颗粒形态以及Ag NOR颗粒分型。结果:①实验后实验组细胞细胞核的面积(P)、周长(At)、最大直径(D),与对照组比较均较低,差异有统计学意义(P〈0.05);②实验后实验组细胞细胞核体密度(Vv)、表面积密度(Sv)、平均体积(V)、平均表面积(S)以及三维形状因子(Rsv),与对照组比较均较低(P〈0.05),实验组核数密度(Nv)明显高于对照组(P〈0.05);③实验后实验组Ag NOR颗粒的直径大于对照组(P〈0.05);实验组Ag NOR颗粒的截面数、面积以及其余和面积的比值均低于对照组(P〈0.05);④实验后实验组单一型、核仁内型Ag NOR颗粒所占的百分比高于对照组,而聚集型、弥散型Ag NOR颗粒所占的百分比低于对照组,差异有统计学意义(χ^2=6.313,P〈0.05)。结论:青蒿琥酯能够使皮肤癌细胞核形态及Ag NOR颗粒形态趋于规则,抑制肿瘤细胞增殖,对临床具有指导意义,值得临床推广。 Objective: To explore the effect of artesunate on the morphology of skin cancer nuclear and Ag NOR particles. Methods: The purchase of human epidermal squamous cell line A431 cells were randomly divided into two groups. After conventional culture,1 m L artesunate with the concentration of 200 mol/L( with 10% fetal bovine serum dilution) was used in the experiment group. Control group was given the equal amount of 10% fetal bovine serum. After 48 h culture,produce the section. Use HE staining and silver staining method. Through color pathological image,analyze the two groups' nuclear morphology,cell structure and morphology of Ag NOR particle and Ag NOR particle classification. Results: 1Area( P),perimeter( At) and maximum diameter( D) in the experiment group after the experiment compared with the control group's were significantly lower( P〈0. 05). 2After the experiment,the experiment group cells' volume density( Vv),surface density( Sv),the average volume( V),the average surface area( S) and 3D shape factor( Rsv) were significantly lower compared with the control group' s( P〈0. 05). While the experiment group's nuclear density( Nv) was significantly higher than the control group's( P〈0. 05). 3After the experiment,the experiment group's diameter of Ag NOR particle was higher than that of control group( P〈0. 05). The experiment group's Ag NOR particles and the rest of sectional area and area ratio were significantly lower than the control group's( P〈0. 05). 4After the experiment,the experiment group's percentage of single type and nucleolus type of Ag NOR particles was higher than that of the control group and the percentage of aggregation type and diffuse type of Ag NOR particles was lower than that of the control group( χ~2= 6. 313,P〈0. 05). Conclusion: Artesunate can cause skin cancer cell nuclear morphology change and the morphology of Ag NOR particle tends to be regular,inhibiting the proliferation of tumor cells. It has the instruction significance to the clinical promotion.
