摘要
目的:免疫细胞化学法检测不同组小鼠含药血清作用对肿瘤组织中PCNA的影响;ELISA方法检测鼠外周血s IL-2R。方法:小鼠自接种瘤株细胞24 h后称重、编号,随机分为8组,每组5只,分别为:1回药爱康方高剂量组(AKH);2荷瘤模型组(对照组);3回药爱康方中剂量组(AKM);4单纯化疗药组(CTX组);5回药爱康方中剂量组加化疗组(AKM+CTX);6回药爱康方低剂量加化疗药组(AKL+CTX);7回药爱康方低剂量组(AKL);8回药爱康方高剂量加化疗药组(AKH+CTX)。结果:与模型组比较高、中、低、高+环磷酰胺、中+环磷酰胺、低+环磷酰胺,组织中PCNA的表达都降低,差异具有显著统计学意义(P<0.05);血清中s IL-2R的表达都降低,差异具有显著统计学意义(P<0.05)。结论:本实验证明回药爱康方高中低剂量以及高中低剂量与环磷酰胺联合用药可以通过调节PCNA以及Sil-2的表达来抑制肿瘤细胞的增殖,对临床使用提供了更为可靠的实验依据。
Objective: By immunocytochemistry method to test the effect on PCNA in tumor tissue of different groups' mice serum. By ELISA method to detect SIL- 2R in the peripheral blood. Methods: After inoculated tumor cell strains 24 h,the mice,weighed and marked the serial number,were randomly divided into 8 groups,each group 5,respectively:( 1) Aikang Formula high dose group( AKH);( 2) tumor- bearing model group( control group);( 3) Aikang Formula middle dose group( AKM);( 4) simple chemotherapy group( CTX);( 5) Aikang Formula middle dose + chemotherapy group( AKM + CTX);( 6) Aikang Formula low dose and chemotherapy group( AKL + CTX);( 7) Aikang Formula low dose group( AKL);( 8) Aikang Formula high dose and chemotherapy group( AKH + CTX). Results: Compared with model group,high,low and high dose group,high dose + cyclophosphamide,middle dose + cyclophosphamide,low dose + cyclophosphamide groups' expressions of PCNA were elevated with significant difference( P〈0. 05). The expressions of serum SIL- 2R were reduced with significant difference( P〈0. 05). Conclusion:This experiment proved that Aikang Formula( high,middle and low dose) and the combination with cyclophosphamide can inhibit the proliferation of tumor cells by regulating the expressions of PCNA and SIL- 2R,providing more reliable experimental basis.
