电针内关穴对心肌缺血ASIC3^(-/-)小鼠瞬时外向钾离子通道相关蛋白表达的影响 被引量:8

摘要 目的:探讨电针内关穴对心肌缺血ASIC3基因敲除(ASIC3^(-/-))小鼠心肌细胞瞬时外向钾离子通道相关蛋白Kv1.4、Kv4.2、Kv4.3及KCh IP2表达的影响。方法:ASIC3^(-/-)小鼠36只,随机分为空白组、模型组、内关组、列缺组、足三里组和非经非穴组,每组6只,皮下注射异丙肾上腺素建立心肌缺血模型。检测小鼠电针治疗前后心电图T波电压的变化,Western blot检测Kv1.4、Kv4.2、Kv4.3和KCh IP2蛋白表达量。结果:治疗后,与模型组相比,内关组、列缺组和足三里组小鼠T波电压和蛋白表达量均有所回升(P<0.05),其中内关组优于列缺组及足三里组(P<0.05);非经非穴组与模型组之间无明显差异(P>0.05)。结论:电针内关穴、列缺穴和足三里穴均可提升心肌缺血ASIC3^(-/-)小鼠心肌细胞瞬时外向钾离子通道相关蛋白的表达,且心包经内关穴的针刺效应优于列缺穴和足三里穴。 Objective: To explore the effect of electro- acupuncture( EA) at Neiguan( PC6) in the protein expressions of Kv1. 4,Kv4. 2,Kv4. 3 and KCh IP2 on the transient outward potassium channels of acid- sensing ion channel 3 gene knockouted( ASIC3^(-/-)) mice with myocardial ischemia( MI). Methods: Thirty ASIC3^(-/-)mice were randomly divided into 6 groups,including control group,modeling group,PC6 group,Lieque( LU7) group,Zusanli( ST36) group and non- point group. The MI model was set up by subcutaneous injection of isoproterenol( ISO) and then measure T- wave voltage before and after treatment. The protein expressions of Kv1. 4,Kv4. 2,Kv4. 3 and KCh IP2 were analyzed by Western Blot. Results: After EA treatment,compared with the modeling group,the T- wave voltage and the protein expressions of PC6 group,LU7 group and ST36 group were obviously increased( P〈0. 05). The protein expression of PC6 group was higher than that of LU7 group and ST36 group( P〈0. 05). There was no significant difference between modeling group and non- point group( P〉0. 05). Conclusion: EA at PC6 and LU7 can increase the protein expression of transient outward potassium channels in myocardial cells and the effect of PC6 was better than that of LU7 and ST36.
