摘要 对新鲜的血液或者血痕,采用直接扩增和chelex-100直接处理提取DNA,一般都能获得完整、均衡的DNA分型,但在日常检案过程中,陈旧血痕、载体潮湿、委托方送检的样本由于保存不当导致血液和少量血痕检材腐败的情况也比较常见。这类检材DNA采用普通的chelex-100直接提取很难获得比较完整的DNA分型。结合一列检案,分别用chelex-100、磁珠法、微量DNA提取试剂盒三种方法专门针对腐败血液进行比较分析。 Usually, complete and balanced DNA typing can be extracted from fresh blood or bloodstains through direct amplification and chelex-100. In case of daily inspection process, however, such circumstances are quiet common, like old blood stains, moist carriers, decayed blood and small amount of decayed bloodstain samples caused by improper preservation. For such samples, it is hard to extract complete DNA typing through the method of chelex-100. Combined with a case, this paper compares and analyses three DNA extraction methods applied to decayed blood: chelex-100, paramagnetic particle method and trace DNA extraction kit.
