鞘内注射黄芪注射液对大鼠局灶性脑缺血后神经干细胞的作用 被引量:3

摘要 目的采用动物实验研究鞘内注射黄芪注射液对局部病灶性的脑缺血后神经干细胞的作用。方法选择健康的雄性SD大鼠60只,随机分为三组,分别为假手术组、模型组和黄芪组,每组大鼠各20例。模型组按照Longa法进行局灶性脑缺血模型的建立,黄芪组在模型建立的基础上在鞘内注射黄芪注射液,假手术组则仅进行手术,但是不进行模型的建立。比较三组大鼠手术后6 h、12 h、24 h和48 h的神经症状评分和脑源性神经营养因子(BDNF)阳性细胞计数,并对组织染色情况进行观察。结果黄芪组在精神、肢体力量、日常活动等方面均好于假手术组和模型组,假手术组在手术后神经症状评分为0,没有神经症状,模型组和黄芪组的神经症状评分均高于假手术组,黄芪组在各时间段的神经症状评分低于模型组,差异具有统计学意义(P<0.05);术后时间越长模型组和黄芪组的神经症状评分越低,差异具有统计学意义(P<0.05)。模型组和黄芪组的BDNF阳性细胞计数均高于假手术组,且黄芪组高于模型组,模型组在术后24 h时BDNF阳性细胞计数最高,黄芪组BDNF阳性细胞计数随手术时间的延长而增加,差异具有统计学意义(P<0.05)。结论黄芪注射液对局灶性脑缺血大鼠的神经功能具有一定的改善作用,能够减轻脑组织的病例变化,促进神经干细胞的增殖,对于缺血性脑梗死具有一定的治疗作用。 Objective To study the effect of intrathecal injection of Astragalus Injection on focal cerebral ischemia neural stem cells in mice. Methods A total of 60 healthy male SD rats were randomly divided into three groups:sham operated group,model group and Astragalus group. 20 cases in each group were randomly divided into groups. In the model group,the establishment of focal cerebral ischemia model was es-tablished by Longa method,and the model was established on the basis of the model. 6 h,12 h,24 h and 48 h positive cell count were compared between the three groups after operation,and the BDNF positive cells were counted. Results Mental status,physical strength,daily activities in Astragalus group were better than those in sham operation group and model group. The neurological symptom score was 0,no symptoms. The scores of model group and Astragalus group were significantly higher than those of sham operation group. BDNF positive cells in model group and Astraga-lus group were higher than those in sham operation group,and the number of BDNF positive cells in model group was higher than that in model group,and the number of BDNF positive cells increased with the increase of operation time,and the difference was statistically significant( P 〈0. 05). Conclusion Astragalus injection can improve the nerve function of rats with cerebral infarction,which can reduce the change of brain tis-sue and promote the proliferation of neural stem cells.
