Twist1通过NF-κB信号通路对人乳头状甲状腺癌上皮-间质转化的调控机制研究 被引量:11

摘要 目的探究Twist1对人乳头状甲状腺癌的侵袭转移的影响及相关蛋白表达的影响。方法利用MTT检测Twist1对人乳头状甲状腺癌乳头状癌细胞(IHH-4)细胞增殖能力的影响;利用转孔小室(Transwell小室)实验检测Twist1对人乳头状甲状腺癌IHH-4细胞侵袭能力的影响;采用免疫印迹试验(Western Blot)检测上皮-间质转化(EMT)相关标志蛋白和核因子(NF-κB)信号通路的分子蛋白的改变情况。结果在人乳头状甲状腺癌IHH-4细胞中沉默Twist1后,细胞的增殖能力没有发生显著变化;通过沉默Twist1可以降低人乳头状甲状腺癌IHH-4细胞侵袭和迁移能力,降低了EMT的恶性程度;Western Blot检测沉默Twist1后EMT相关蛋白上皮钙黏蛋白(E-Cadherin)、神经钙黏蛋白(N-cadherin)、波形蛋白(Vimentin)和NF-κB信号通路的p-NF-κB P65均发生明显改变。结论在人乳头状甲状腺癌中Twist1起到癌基因的作用,并且通过NF-κB信号通路对人乳头状甲状腺癌上皮-间质转化起到促进作用,为临床治疗人乳头状甲状腺癌提供科学依据及新的治疗靶点和思路。 Objective To investigate the relationships among Twistl, EMT and 出e invasion and metastasis of papillary thyroid carcinoma.Methods We screened the proliferation inhibition of Twistl on IHH -4 cells by using MTT. We detected the invasive ability of Twistl onIHH -4 cells by using transwell. We detected EMT related proteins using Western Blot. Results The silence of Twistl did not show significantinhibitory effect on the proliferation of IHH - 4 cells significantly. The silence of Twistl showed significantly inhibitory effect on the invasion ofIHH - 4 cells significantly. Western Blot test EMT related proteins E - Cadheri, N - cadherin, Vimentin and p - NF - kBP65 which were obviouslychanged. Conclusion Twistl regulates the epithelial - mesenchymal transition via the NF - KB pathway in papillary thyroid carcinoma.
