摘要 研究了碱性蛋白酶和木瓜蛋白酶双酶法水解蚕蛹蛋白的工艺条件。通过单因素试验探讨了温度、加酶量、底物浓度、水解时间、酶质量比(碱性蛋白酶∶木瓜蛋白酶)对蚕蛹蛋白水解效果的影响,然后通过正交试验优化水解工艺务件。结果表明,影响水解率的主次关系依次为:温度、底物浓度、时间、酶用量、酶比。优化的工艺条件为:温度55℃,加酶量3.0%,底物浓度3%,水解时间6 h,酶质量比4∶1,上述条件下蚕蛹蛋白水解率达25.04%。 The technology for silkworm pupa protein hydrolysis by double enzymatic(alkaline protease and pa-pain) was studied.Single factor and orthogonal experiments were used to study the effects of the parameters such as temperature,amount of enzyme,substrate concentration,time,mass ratio of a1kaline protease and papain on the hydrolysis degree of silkworm pupa protein. The results showed that the temperature was the more important factor that affected the effect of hydrolysis,following substrate concentration and time and amount of enzyme and enzyme mass ratio, the optimal conditions were obtained with orthogonal experiments, as follows:the tempera-ture was 55℃, the amount of enzyme was 3.0%, the substrate concentration was 3%, the time was 6 h and the enzyme mass ratio was 4∶1. The hydrolysis degree of silkworm pupa protein under these conditions could be 25.04%.
