摘要
利用紫外分光光度计,采用苯酚-硫酸法,以葡萄糖为标准品,对比苯酚-硫酸法(方法Ⅰ)、《中国药典》中方法(方法Ⅱ)及改进后的苯酚-硫酸法(方法Ⅲ),在485 nm处测定枸杞多糖的含量,比较3种方法的优劣。试验表明:3种方法标准曲线相关系数分别为0.930 9、0.991 8、0.996 8;吸光值稳定性RSD值分别为10.95%、2.13%、0.13%;回收率试验中方法Ⅲ的相对误差较小,无显著差异。可见方法Ⅲ最优。
Using the phenol-sulfuric acid method, with glucose as the standard by UV spectrophotometer and comparison of phenol-sulfuric acid method (method I ), the method in China Pharmacopoeia (method Ⅱ ) and the method improved (method Ⅱ),the content ofLycium barbarum polysaccharide was detected under 485 nm to compare the advantages of the three methods. Experiments showed that the three methods standard curve cor- relation coefficient were 0.930 9,0.991 8, 0.996 8, RSD of light absorption value stability were 10.95 %,2.13 %, 0.13 % ;the relative error of recovery rate of method Ⅲ was small, which showed no significant difference. It meant that the method III was the best.
