摘要
目的研究通窍活血汤对局灶性缺血性脑卒中(MCAO)神经损伤的影响。方法选择50只SPF级健康Wistar雄性大鼠,用随机数表法分为5组:假手术组、模型组、对照组和低、高2个剂量实验组每组10只。除对照组外,其余4组大鼠均制作MCAO模型,低、高2个剂量实验组,分别给予0.9,1.8 g·m L^(-1)通窍活血汤5 m L·kg^(-1)灌胃;对照组给予36 mg·m L^(-1)吡拉西坦片水溶液5 m L·kg^(-1)灌胃;假手术组和模型组灌服5 m L·kg^(-1) 0.9%NaCl,均每日灌胃2次,共20 d。观察5组大鼠的神经运动功能、血清一氧化氮(NO)、血浆内皮素- 1(ET- 1)和两者比值NO/ET- 1的水平以及脑组织谷氨酸(Glu)、天冬氨酸(Asp)水平变化情况。结果给药后20 d,低、高2个剂量实验组运动功能评分为(6.15±0.08),(6.58±0.08)分,对照组为(6.08±0.15)分,这3组的运动功能评分均较模型组的(5.07±0.12)分明显升高,组间比较差异有统计学意义(P<0.05)。低、高2个剂量实验组血浆ET- 1含量为(69.72±5.76),(57.32±6.14)pg·m L^(-1)和对照组为(70.12±5.86)pg·m L^(-1),这3组均明显低于模型组的(94.22±6.38)pg·m L^(-1)(P<0.05)。低、高2个剂量实验组NO水平为(51.82±2.36),(65.72±2.44)μmol·L^(-1)和对照组为(52.68±2.55)μmol·L^(-1),这3组的NO水平明显高于模型组(40.36±2.48)μmol·L^(-1),组间比较差异有统计学意义(P<0.05)。2个剂量实验组Glu水平为(6.35±1.18),(5.08±1.06)μmol·L^(-1)和对照组为(6.68±0.94),这3组的Glu水平明显低于模型组的(9.58±1.34)μmol·L^(-1),组间比较差异有统计学意义(P<0.05)。2个剂量实验组Asp水平为(10.16±0.12),(8.24±0.28)μmol·L^(-1)和对照组为(10.04±0.18)μmol·L^(-1),这3组的Asp水平明显低于模型组(12.42±0.25)μmol·L^(-1),组间比较差异有统计学意义(P<0.05)。结论高剂量通窍活血汤可明显改善缺血性脑卒中大鼠神经运动功能,提高血清NO、ET- 1水平,降低脑组织Glu、Asp水平。
Objective To study effects of Tongqiao Huoxue Decoction( THD) on ischemic stroke nerve injury in rats. Methods The 50 SPF level healthy male Wistar rats was selected using a random number table method and divided into sham group,model group,low and high doses of two test groups,and control group,ten rats in each group. The focal cerebral ischemia( MCAO) model were prepared. Low and high doses of two test groups were given THD 0. 9,1. 8 g·m L^-1in accordance with 5m L·kg^-1orally,control group rats were given 36 mg·m L^-1piracetamtablets aqueous solution 5 m L·kg^-1orally; sham group and model group were fed with 5 m L·kg^-1saline. All rats were fed twice a day,a total of 20 d. The neuromotor function,changes level of serum nitric monoxide( NO),plasma endothelin-1( ET-1) and the ratio between the NO / ET-1 levels and brain glutamic acid( Glu),aspartic acid( Asp) were observed in rats of five groups. Results After administration of 20 d,the motor function scores in low and high doses of two test groups with( 6. 15 ± 0. 08),( 6. 58 ± 0. 08) and control group with( 6. 08 ± 0. 15),were significantly higher in model group with( 5. 07 ± 0. 12)( P〈0. 05). The ET-1 level in low and high doses of two test groups with( 69. 72 ± 5. 76),( 57. 32 ± 6. 14) pg·m L^-1and control group with( 70. 12 ± 5. 86) pg·m L^-1,were significantly lower in model group with( 94. 22 ± 6. 38) pg·m L^-1( P〈0. 05). The serum NO level in low and high doses of two test groups with( 51. 82 ± 2. 36),( 65. 72 ± 2. 44) μmol·L^-1and control group with( 52. 68 ± 2. 55) μmol·L^-1,was increased signi-ficantly than in model group with( 40. 36 ± 2. 48) μmol·L^-1( P〈0. 05). The levels of Glu in low and high doses of two test groups with( 6. 35 ± 1. 18),( 5. 08 ± 1. 06) μmol · L^-1and control group with( 6. 68 ± 0. 94) μmol·L^-1were decreased significantly than in model group with( 9. 58 ± 1. 34) μmol·L^-1( P〈0. 05).The levels of Asp in low and high doses of two test groups with( 10. 16 ± 0. 12),( 8. 24 ± 0. 28) μmol·L^-1and control group with( 10. 04 ± 0. 18) μmol·L^-1was decreased significantly than in model group with( 12. 42 ± 0. 25) μmol·L^-1( P〈0. 05). Conclusion High dose THD can obviously improve nerve motor function of the ischemic cerebral apoplexy rats,increase the serum and plasma NO and ET 1 level,while lower Glu,Asp levels in brain tissue.
