摘要
目的:研究富血小板纤维蛋白(PRF)及其所含三因子TGF-β1、PDGF-AB、VEGF分别对大鼠脂肪干细胞(ADSCs)增殖和黏附的影响。方法:将ADSCs培养在含有PRF膜和不同浓度的PDFG-AB、TGF-β1、BEGF中,细胞黏附实验检测培养2 h后细胞黏附能力,CCK-8法检测培养1~7 d细胞增殖。结果:黏附实验显示,PRF组的黏附细胞数显著高于阴性对照组(P〈0.05);不同浓度的PDGF-AB组内黏附细胞数的差异无统计学意义(P〉0.05);不同浓度的TGF-β1、VEGF组内黏附细胞数的差异有统计学意义(P〈0.05)。CCK-8实验表明,PRF组的细胞增殖在各个时间点(1、3、5、7 d)显著高于阴性对照组(P〈0.05);不同浓度的PDGF-AB、VEGF组内细胞的增殖差异有显著性(P〈0.05);不同浓度的TGF-β1组细胞增殖的差异无显著性(P〉0.05)。结论:PRF能提高ADSCs的增殖和黏附的能力。PDGF-AB和VEGF可促进ADSCs的增殖;TGF-β1和VEGF均可促进ADSCs的黏附,并呈一定的量效正相关。
Objective: To study the effects of PRF and recombinant h PDGF-AB,TGF-β1 and VEGF on the proliferation and adhesion of rat adipose tissue-derived stem cells( ADSCs) in vitro. Methods: ADSCs were cultured with PRF membrane and various doses of PDGF-AB,TGF-β1 and VEGF,cell adhesion was examined by adhesion assay after 2 culture,cell proliferation was examined by CCK-8 kit after 1- 7 d culture. Results: Cell adhesion assay showed that the adhesive numbers of rat ADSCs in PRF group were significantly higher than those in the negative group( P〈0. 05). The adhesive numbers of the ADSCs treated by PDGF-AB showed no significantly difference among different concentration groups( P〉0. 05). The adhesive numbers of the ADSCs treated by VEGF or TGF-β1 at different concentrations showed significant difference( P〈0. 05). CCK-8 kit assay showed that at different time points,the A values of ADSCs in PRF group were significantly higher than those of the negative control group( P〈0. 05). The A values of ADSCs in VEGF or PDGF-AB groups at different concentrations showed significant difference( P〈0. 05). The A values of rat ADSCs in TGF-β1 group at different concentrations were lower than those in the negative control group( P〈0. 05). Conclusion: PRF as a combination of growth factors may stimulate the proliferation and adhesion of rat ADSCs in vitro. PDGF-AB and VEGF may stimulate the proliferation of rat ADSCs. TGF-β1 and VEGF may stimulate the adhesion of rat ADSCs in a dose-response manner to some degree.
