摘要
目的:优选代谢综合征方水提部分回流提取工艺。方法:采用正交试验法,以黄芪甲苷、毛蕊异黄酮葡萄糖苷、金丝桃苷和蒙花苷的含量做为综合评价指标,考察溶剂用量、提取时间及提取次数对代谢综合征方提取工艺的影响。采用高效液相测定提取液中黄芪甲苷、毛蕊异黄酮葡萄糖苷、金丝桃苷和蒙花苷的含量。结果:提取次数对代谢综合征方水提部分提取效果具有显著性影响,加水量和提取时间对提取效果均无显著影响。故代谢综合征方的最佳水提取工艺为12倍水回流提取3次,每次2.5 h,所得黄芪甲苷、毛蕊异黄酮葡萄糖苷、金丝桃苷和蒙花苷的平均提取量为1.235 mg·g^(-1)、0.580 mg·g^(-1)、3.693 mg·g^(-1)、15.010 mg·g^(-1)。结论:该优选工艺稳定、合理,可为代谢综合征方的大规模生产提供实验依据。
Objective: To optimize the reflux extraction technology of the water extracts of prescriptions for metabolic syndrome.Methods: Orthogonal test method was applied and the contents of Astragaloside,mullein isoflavone glucoside,Hyperoside and Linarin were used as the comprehensive evaluation index to examine the effect of the the amount of solvent,extraction time and extraction times on the extraction process of prescriptions for metabolic syndrome. High performance liquid was used to measure the content of Astragaloside,mullein isoflavone glucoside,Hyperoside and Linarin liquid extracts. Results: The frequency of extracting had significant effect on water extracts of prescriptions for metabolic syndrome,while the amount of added water and the extraction time had no significant effect. Therefore,the best extraction technology of the water extracts of prescriptions for metabolic syndrome is three times of extraction with 12 times of water and 2. 5 h each time. The average extraction amount of Astragaloside,mullein isoflavone glucoside,Hyperoside and Linarin was 1. 235 mg·g^(-1),0. 580 mg·g^(-1),3. 693 mg·g^(-1),15. 010 mg·g^(-1)respectively. Conclusion: The extraction technology is stable and reasonable,and it can provide laboratory data for the production of prescriptions for metabolic syndrome.
