一种稀土荧光化合物的合成及光谱性质研究

摘要 目的研制新型时间分辨荧光免疫分析螯合剂。方法以4,4′-二溴-6,6′-二溴甲基-2,2′-联吡啶为起始反应原料设计合成出一种新型化合物4,4′-二(对氨基苯乙炔基)-6,6′-二(N,N-二(叔丁氧基羰甲基)氨甲基)-2,2′-联吡啶,并对其结构进行确认。结果将该化合物与铕离子结合形成的稀土荧光化合物的荧光性质进行研究,表明其主要发射波长为595 nm(~5D_0-~7F_1)、618nm(~5D_0-~7F_2),荧光寿命为643μs。当荧光溶液放置72h,荧光强度值无明显变化。在该荧光体系下,Eu^(3+)浓度分析检测范围为10^(-5)~10^(-11)mol/L。结论该荧光化合物结构有较好的稳定性,为固相时间分辨免疫体系奠定了基础,具有广泛的应用前景。 Objective To develop new types of solid-phase time-resolved fluorescence immunoassay chelating agents. Metheds A new compound 4,4'-di(p-aminophenylethynyl)-6,6'-bis[N,N-bis(carboxymethyl) aminomethyl]-2,2'-bipyridine was synthesized with 4,4'-dibromo-6,6'-bis(bromomethyl)-2,2'-bipyridine. Compounds with europium ions to form fluorescent complexes of rare earth, analysis the spectral properties of the europium complexes. Results The main emission wavelength of the fluorescent complex was 618nm and the fluorescence lifetime was 673μs when the concentration was 1×10-5mol·L-1. The detection range of europium ions is between 1×10^-5 mol·L^-1 and 1×10^-11mol·L^-1. Fluorescence of the fluorescent compound has little influence after 72h. Conclusion The complex has high sensitivity and strong stability. It builds up a good basis for soild-phase time-resolved fluorescence in the future and it has an applicable prospect.
