番茄抗黄化曲叶病ty-5基因的PCR快速检测 被引量:1

摘要 以番茄抗病材料CLN32120a-23(含ty-5基因)和感病材料Moneymaker(不含抗病基因)为试材,通过杂交、自交获得的F1、F2代,利用SSR分子标记技术筛选出了1个与抗病性状共分离的共显性SSR标记。结果表明:该标记在CLN32120a-23中可以扩增出550bp的条带,在Moneymaker中可以扩增出780bp的条带,在F1中可以扩增出上述2条条带,标记结果与田间鉴定基本一致,证明该标记能够区分抗病材料、感病材料及杂合抗病材料,是与番茄抗黄化曲叶病毒病基因ty-5紧密连锁的共显性标记。对27份番茄材料进行检测,鉴定出有15份材料基因型为ty-5/ty-5,分子标记检测与田间检测结果吻合率达80%。该标记可用于番茄抗黄化曲叶病毒ty-5基因的PCR快速检测,同时为ty-5分子标记辅助选择育种的应用提供一定的参考依据。 Taking CLN32120a-23(resistant materials)and Moneymaker(susceptible materials)as experimental materials which is resistant to yellow leaf curl virus in tomato,F1 .F2 were obtained through hybridization, selfcross, using SSR technology,a molecular marker was obtained. The results showed that SSR marker had a 550 bp amplification fragment in homozygous resistant lines,a 780 bp fragment in susceptible lines and both 780 bp and 550 bp fragments in heterozygous resistant lines. The detection results were consistent with those of field identification. So the SSR marker,tightly linked to ty-5 gene,could distinguish homozygous and heterozygous resistant lines and susceptible lines. 27 tomato materials individuals were detected. There were 15 homozygous resistant lines,the molecular detection was 80% coincident with the results of field identification. The results showed that ty-5 SSR marker could be used for rapid identification of ty-5 resistant gene in tomato.
