肺纤维化前期肺组织中ICAM-1、TGF-β、CTGF的表达及其镜下形态改变的动态观察 被引量:9

摘要 目的动态观察博莱霉素诱发大鼠肺纤维化模型从肺损伤到肺纤维化前期肺组织中细胞间黏附分子-1(intercellular cell adhesion molecule-1,ICAM-1)、转化生长因子-β(transforming growth factor-β,TGF-β)、结缔组织生长因子(connective tissue growth factor,CTGF)水平的变化,探讨肺纤维化前期的发病机制。方法雌性SD大鼠108只,随机分为肺纤维化模型组、内毒素组、空白对照组(每组36只),并于干预后第3、7、14天取各组肺组织进行免疫组化SABC法观察。结果造模后各组TGF-β、ICAM-1、CTGF的表达依次为空白对照组<内毒素组<肺纤维化模型组。肺纤维化模型组显著高于空白对照组及内毒素组(P<0.05)。光镜下观察空白对照组中肺组织结构正常;肺纤维化模型组第7天未见明显肺纤维化改变,第14天VG染色可见少量胶原纤维形成;内毒素组第7天有轻度肺泡炎,第14天肺泡炎消失。结论肺纤维化前期纤维化的发生与TGF-β、ICAM-1、CTGF三种细胞因子均有密切关系,三者可能起协同作用。 Purpose To dynamically observe the level of cytokines intercellular cell adhesion molecule-I ( ICAM-I ), transforming growth factor-β (TGF-β) , connective tissue growth factor (CTGF) changes in the bleomycin-induced pulmonary fibrosis rats that from lung injury to the early stage of pulmonary fibrosis, and to explore the early pathogenesis of pulmonary fibrosis. Methods 108 cases of female SD rats were randomly divided into bleomycin group (model group) , endotoxin group and the normal group. The lung tissues from each group who was intervened at 3,7, 14 days were taken to perform immunohistochemical SABC method. Results The expression of the TGF-β, ICAM-1, CTGF at 3, 7, 14 day after modeling was from low to high as follows : normal group 〈 endotoxin group 〈 model group. Model group was significantly higher than the normal group and endotoxin group ( P 〈 0. 05 ). Microscopically, The control group had normal lung tissue structure. In the model group, there was no significant change at 7th day, VG staining showed a small amount of collagen fibers at 14th day. In the endotoxin group, there was mild alveolitis and a small amount of neutrophil infiltration at 7th day, but alveolitis disappeared at 14th day. Conclusion TGF-β, ICAM-1, CTGF may be closely related with the early fibrosis of pulmonary fibrosis, which suggest these three cytokines may play a synergistic effect.
