摘要
目的探讨槐角染料木素对哮喘小鼠白介素-5(IL-5)、白介素-13(IL-13)含量的影响。方法 32只BALB/c雌性小鼠随机分为对照组(n=10)、模型组(n=11)和干预组(n=11)。模型组和干预组采用卵清蛋白(ovalbumin,OVA)腹腔注射致敏和雾化吸入激发,建立哮喘小鼠模型。干预组于激发前腹腔注射染料木素30 mg/kg。观察小鼠哮喘症状的发生情况,检测小鼠血清OVA特异性IgE水平。检测肺泡灌洗液(bronchoalveolar lavage fluid,BALF)中嗜酸性粒细胞(eosinophil,EOS)计数及其分类比值和细胞因子IL-5、IL-13含量。结果与模型组相比,干预组小鼠哮喘症状出现较迟,且症状较轻微。与对照组相比,模型组BALF中EOS计数及分类比值、IL-5和IL-13的含量及血清中OVA特异性Ig E水平均明显增加(均P<0.01)。与模型组相比,干预组BALF中EOS计数及分类比值、IL-5和IL-13的含量及血清中OVA特异性IgE水平均明显降低(均P<0.01)。结论染料木素可缓解小鼠哮喘症状,可能与其降低哮喘小鼠IgE水平,降低IL-5及IL-13的含量,从而减轻气道炎症有关。
Objective To investigate the effect of genistein of fructus sophorae on the contents of interleukin-5(IL-5)and interleukin-13(IL-13) in mice with asthma. Methods 32 BALB/c female mice were divided into the control group(n=10), model group(n=11) and intervention group(n=11) randomly. Model group and intervention group were sensitized by intraperitoneal injection with ovalbumin(OVA) and excited by aerosol inhalation with OVA. The intervention group was given intraperitoneal injection with 30 mg/kg genistein before excitation. The asthma symptoms of mice were observed. The serum levels of anti-OVA Ig E, the counts and classification ratios of eosinophils(EOS), the contents of IL-5 and IL-13 in bronchoalveolar lavage fluid(BALF) were detected. Results Compared with the model group, the asthma symptoms of the intervention group were appeared late and mildly. Compared with the control group, the counts and classification ratios of EOS, the contents of IL-5 and IL-13 in BALF, the serum levels of anti-OVA IgE in the model group were significantly increased(all P 0.01). Compared with the model group, the counts and classification ratios of EOS, the contents of IL-5 and IL-13 in BALF, the serum levels of anti-OVA IgE in the intervention group were reduced significantly(all P 0.01). Conclusion Genistein can relieve the symptoms of asthma, may concern with it's reduction the levels of Ig E, the contents of IL-5 and IL-13, and inhibition airway inflammation.
