海藻糖合成酶诱导条件优化研究

摘要 采用乳糖诱导大肠杆菌产海藻糖合成酶,用以转化麦芽糖为海藻糖,利用高效液相色谱法测定海藻糖的转化率。采用单因素试验和正交试验研究乳糖诱导时的pH、温度、转速和持续时间四个因素对海藻糖转化率的影响,寻找大肠杆菌产海藻糖合成酶转化海藻糖的最优条件组合,为海藻糖工业化生产提供参考依据。结果表明,诱导温度和持续时间对海藻糖转化率的影响较大,海藻糖合成酶最佳诱导条件为:pH 7.4、温度26℃、转速500 r/min、诱导时间10 h。在最佳条件下,海藻糖的转化率可达69.45%。 Trehalose synthase was produced by Escherichia coli when induced by lactose, which could convert maltose into trehalose, and the transformation efficiency of trehalose synthase was confirmed using the method of high performance liquid chromatography. For confirming the optimal condition of the transformation efficiency oftrehalose synthase from Escherichia coli, inducting the pH, temperature, rotation rate and time was researched with methods of single factor and orthogonal tests, which could provide reference for the industrial production of trehalose. The results showed that the effect of transformation time and induction temperature on conversion rate was higher. The best collocation was as follows: pH 7.4, induction temperature 26 ℃, speed 500 r/min, induction time 10 h. The conversion rate of trehalose was as high as 69.45% trader the best collocation.
