竹节蓼石油醚部位抗肿瘤活性组分的筛选 被引量:4

Screening Anti-tumor Components in Petroleum Ether Extracts from Homalocladium platycladum

摘要 对竹节蓼石油醚部位分离所得组分进行了体外抗肿瘤活性筛选。MTT法研究硅胶吸附层析分离所得组分对肿瘤细胞株PC-3、A549增殖的影响,相差显微镜观察细胞形态变化,DAPI染色法检测细胞凋亡形态学,薄层色谱结合理化性质分析有效组分中的成分,波谱数据鉴定结构。结果显示分离所得的5种组分中,C_(6-8)对PC-3有增殖抑制作用;C21-24、C_(25-28)对PC-3、A549有较强的增殖抑制作用,且呈量效关系,作用后细胞形态发生了明显改变,DAPI染色结果显示二者都促进了肿瘤细胞的凋亡;薄层色谱分析出有效组分中共含5种化合物,其中单体C7鉴定为羽扇豆醇;C_(21-24)、C_(25-28)组分初步鉴定为甾体和三萜类化合物。 The object of this study was to screen the anti-tumor active fraction of petroleum ether extracts from Homalo- cladium platycladum in v/tro. MTr assay was used to study the anti-tumor efficacy of the various fractions isolated from the petroleum ether extracts against tumor cell lines PC-3 and A549. Phase contrast microscopy was used to observe the cell morphological changes after treated with the active fractions. DAPI staining method was used to determine the apep- tosis of PC-3 and A549 ceils induced by the active fractions. Thin layer chromatography combined with physical and chemical properties analysis was used to detect components in the active fractions, NMR analysis was used to identify the structure of monomer. Results indicated that the C21a4 ,Czs.2s of the 5 separated fractions was the anti-tumor active frac- tions,which significantly inhibited the growth of PC-3 and A549 with dose and time-dependent manner,C6,s only had in- hibitory effect on the proliferation of PC-3. Phase contrast microscopy observation also showed the two fractions can sig- nificantly alter the cell morphology and confirmed the reliability of the MTT results. DAPI staining results showed that C21-24 and C25-28 induced cell apeptosis of PC-3 and A549. TLC results showed that there have 5 components in the 3 ac- tive fractions,C7 isolated from C6.swas a monomer and identified as lupeol;C21-24 and C25-28 was identified as steroid and triterpenoid compounds.
