摘要
目的本实验以吲哚胺2,3双加氧酶(IDO)为切入点,对比分析IDO-si RNA干扰前后胃癌细胞株MGC803中IDO及IL-6表达水平变化,研究IDO在胃癌及抑郁中发生、发展的作用,探讨胃癌促发抑郁的可能分子机制。方法采用si RNA干扰技术转染胃癌细胞株MGC803,免疫细胞化学法及RT-PCR法观察IDO蛋白及基因水平的变化,ELISA法检测转染si RNA前后胃癌细胞株上清液中IL-6表达水平,研究IDO对胃癌细胞株以及抑郁相关细胞因子的影响。结果胃癌细胞MGC803经转染IDO-si RNA后IDO的表达经免疫细胞化学及RTPCR法的检测均明显下降(P﹤0.05);转染IDO-si RNA 24 h后胃癌细胞MGC803的细胞上清液中IL-6的表达较未转染前明显降低,差异有统计学意义(P﹤0.01)。结论 IDO在胃癌细胞株MGC803中高表达;IDO-si RNA可抑制胃癌细胞株MGC803中IDO的表达;胃癌细胞株MGC803经IDO-si RNA干扰后分泌的细胞因子IL-6的表达减弱,表明IDO与IL-6的表达存在关联,IL-6可能在胃癌及抑郁的发生发展中起一定的作用。
Objective To observe the expression levels of indoleamine 2, 3-dioxygenase (IDO) and the secretion of interleukin-6 (IL-6) in gastric cancer cell line MGC803 before and after the application of IDO-siRNA, to investigate the effect of IDO in the development of gastric cancer and cancer related depression. Method Gastric cancer cell line MGC803 was transfected with siRNA, the expression changes of IDO-siRNA and IDO protein were determined by RT-PCR and immunocytochemistry. The change of IL-6 secretion was measured by ELISA. The effect of IDO on cancer cell line and depression related cytokines was investigated. Result After application of IDO-siRNA, the expression of IDO and the secretion of IL-6 in the gastric cancer cell line MGC803 was decreased significantly (P〈0.01). Conclusion IDO is highly expressed in gastric cancer cell line MGC803;IDO-siRNA can inhibit the expression of IDO in gastric cancer cell line MGC803:after application of IDO-siRNA, the secretion of IL-6 was decreased significantly in gastric cancer cell line MGC803, indicating that IDO expression and IL-6 was positively correlated, and IL-6 might be engaged in the devel-opment of gastric cancer and related depression.
