尾巨桉3个无性系外植体诱导技术研究 被引量:1

摘要 [目的]研究尾巨桉3个无性系的外植体诱导相关技术。[方法]筛选最佳采集季节、最优取材部位、最佳消毒时间和最优植物生长调节剂(6-BA)浓度,提高尾巨桉无性系外植体的萌芽率。[结果]3个无性系最佳诱导季节为春秋两季;外植体诱导取材的最佳部位是半木质化的枝条;DH32-26、DH32-28外植体的最佳消毒时间为7 min、DH32-29外植体的最佳消毒时间为5 min;适宜外植体诱导的植物生长调节剂组合为6-BA 1.0 mg/L+NAA 0.4 mg/L,此时诱导萌芽率高。[结论]要提高外植体诱导的成功率,需从采集季节、取材部位、消毒时间以及植物生长调节剂组合方面综合考虑,该研究为尾巨桉苗木快速繁殖提供了理论依据。 Objective The aim was to study explant induction relevant technology of Eucalyptus urophylla ×E.grandis three clones.[Meth-od] The optimal collection season, sampling part, disinfection time and concentration of plant growth regulator(6-BA) was selected, so as to im-prove germination rate of explants of Eucalyptus urophylla × E.grandis clones.[ Result] The spring and autumn were the optimal seasons for the explant induction, semi-lignified branches was the most suitable part of explant sampling , the most appropriate disinfection duration with mer-curic chloride is 7 minutes for DH32-26 and DH32-28, and 5 minutes for DH32-29.6-BA1.0 mg/L and NAA0.4 mg/L as plant growth regulator combination was good for explant induction which with the best germination rate.[Conclusion] In order to improve the successful rate of explant induction, a comprehensive consideration should be made in terms of the collection season, sampling part, disinfection time and plant growth reg-ulator combination.The study can provide theoretical basis for rapid propagation of Eucalyptus urophylla × E.grandis seedlings.
