摘要
目的:探讨雷公藤甲素(TP)对胶原诱导关节炎(CIA)大鼠血清和踝关节中MIP-1α、Eotaxin及MCP-1的影响。方法:建立CIA大鼠模型,用不同浓度的TP进行灌胃治疗,治疗期间,对大鼠的足趾厚度和宽度进行测量。治疗3周以后,将大鼠处死,收集血清标本,用ELISA法检测血清中MIP-1α、Eotaxin及MCP-1的表达水平;同时收集关节标本,采用RT-PCR法对大鼠踝关节滑囊组织的MIP-1α、Eotaxin及MCP-1的m RNA表达进行检测;采用western blot对踝关节滑膜组织的MIP-1α、Eotaxin及MCP-1蛋白表达进行检测。结果:与正常组相比,模型组大鼠的关节肿胀程度明显加重;与模型组相比,治疗组大鼠关节肿胀程度明显减轻。与正常组相比,模型组大鼠血清及踝关节中MIP-1α、Eotaxin及MCP-1的表达明显增加(P<0.05);与模型组相比,各用药组大鼠血浆及踝关节滑膜组织的MIP-1α、Eotaxin和MCP-1表达水平较明显降低(P<0.05)。结论:TP能通过抑制CIA大鼠MIP-1α、Eotaxin和MCP-1的表达而发挥治疗关节炎的作用。
This study aimed to investigate the effects of triptolide(TP) on the expressions of macrophage inflammatory protein-1α(MIP-1α), eotaxin and monocyte chemoattractant protein-1(MCP-1) in collagen induced arthritis(CIA) rats. CIA rat model was established successfully and oral administered with different concentrations of TP for 3 weeks. The degree of ankle swelling was measured during the treatment. After treatment, blood samples from all groups were collected for assessing the concentrations of MIP-1α, eotaxin and MCP-1 by ELISA. MIP-1α, eotaxin and MCP-1 m RNA and protein expressions of rats' ankle joints were detected by RT-PCR and western blot, respectively. Our results showed that the degree of ankle swelling of the model group was aggravated compared with the control group, while that of the treatment group was significantly relieved compared with the model group(P〈0.05). The levels of both serum and articular MIP-1α, eotaxin and MCP-1 of the model group were obviously higher than those of the control group(P〈0.05), while the expressions of MIP-1α, eotaxin and MCP-1 from blood and ankle joint of all treated groups were significantly decreased compared with model group(P〈0.05). In conclusion, it was demonstrated that TP downregulated the expressions of MIP-1α, eotaxin and MCP-1 in CIA rats.
