补肾活血方对小鼠HOXA10基因表达水平的影响

Effect of Bushen Huoxue Decoction on Expression of HOXA10 Gene in Mice

目的:观察补肾活血方对小鼠子宫内膜HOXA10 mRNA表达水平的影响,探讨其对子宫内膜容受性的改善作用。方法:将昆明种小鼠随机分为空白组(生理盐水)、模型组(腹腔注射GnRH-α后依次注射HMG、HCG,制造控制性超排卵模型)和中药组(造模同时给予补肾活血方汤剂),比较各组小鼠子宫指数,通过原位杂交技术检测子宫内膜中HOXA10 mRNA的表达水平。结果:1)模型组小鼠与空白组相比,子宫指数明显降低,中药组子宫指数显著高于模型组(P<0.01);2)原位杂交实验结果显示:模型组小鼠子宫内膜HOXA10 mRNA表达水平明显低于空白组,中药组HOXA10表达水平则明显高于模型组。结论:补肾活血方可明显改善超排卵小鼠子宫指数,并显著增加子宫内膜HOXA10表达水平,从而改善子宫内膜容受性,提高临床体外受精-胚胎移植(IVF-ET)的着床率。

Objective ：To observe the effect of Bushen Huoxue prescription on endometrial receptivity in mice and to explore its mechanism. Methods ：60 female non- pregnant mice were weighed and numbered and they were randomly divided into 3 groups ： a blank group （ normal saline group）, a model group （ GnRH - ot ＋ HMG ＋ HCG controlled ovarian hyper stimulation program ） and a traditional Chinese medicine group （GnRH-or ＋ HMG ＋ HCG ＋ Bushen Huoxue decoction）. The uterine index of the mice in each group was compared. The expression level of HOXA10 in endometrium was detected by in - situ hybridization. Results： Compared with that in the blank group, the uterus index of the model group was significantly lower; the uter- ine index of the traditional Chinese medicine group was significantly higher than that of the model group （P 〈 0.01 ）. In - situ hybridization results showed that the expression of HOXA10 in endometrium of mice in model group was significantly lower than that of the control group, and the expression level of HOXA10 in Chinese medicine group was significantly improve significantly higher than that of model group. uterus index of superovulation mice, and can Conclusion：Bushen Huoxue decoction can increase the endometrial HOXA10 expres- sion level obviously, so as to improve the endometrial receptivity, increase the rate of clinical in - vitro fertili- zation and embryo transfer （IVF- ET） implantation.
