摘要
以从传统发酵酸驼乳中优势菌种粪肠球菌作为浓缩型酸驼乳发酵剂的原料菌,制备粪肠球菌液芯微胶囊,以液芯微胶囊作为微反应器,将乳清粉作为基础培养基,补充适当碳氮源、促生长因子、p H缓冲剂通过单因素实验和响应曲面实验优化高密度发酵培养的培养基配方。微囊化粪肠球菌的最佳培养基配方:乳清粉60 g/L,蔗糖18.22 g/L,蛋白胨35.94 g/L,胡萝卜汁152.82 m L/L,CaCO_39.21 g/L,MgSO_40.28 g/L,MnSO_40.18 g/L。按照此配方进行增殖培养时,粪肠球菌的菌体浓度可达到2.1×10^(14)cfu/g。为浓缩型酸驼乳发酵剂提供依据。
In order to get high -density cultural ,the dominant bacteria Enterococcus faecalis isolated from traditional fermented camel milk was studied as initial bacteria under different cultural conditions to make liquid -core microcapsules.Used whey powder as basal medium,the appropriate carbon and nitrogen sources,somatomedin, pH buffers was carried out on the basis of single -factor experiments, and the optimal fermentation medium formula was determined by Response surface methodology.The best cultural medium for microencapsulatated Enterococcus faecalis was designed as follow: whey powder 60 g/L, sucrose 18.22 g/L, peptone 35.94 g/L, carrot juice ]52.82 ml/L, CaCO3 9.21 g/L, MgSO4 0.28 g/L, MnSO4 0.18 g/L. Under above - mentioned recipe, the viable cell concentration reached to 2.1×10^14cfu/g.
