低分子量透明质酸对小鼠巨噬细胞和T细胞的免疫调节作用 被引量:2

摘要 低分子量透明质酸(LMWHA)由自由基氧化降解方法制备,通过体内动物实验和体外细胞实验研究了LMWHA对小鼠巨噬细胞和T细胞的免疫调节活性。在体内免疫抑制鼠实验中,察看了LMWHA对2,4-二硝基氟苯(DNFB)诱导的迟发型超敏反应(DTH)的影响,测定了血清溶菌酶含量、脾脏以及胸腺指数。结果表明:LMWHA(20~120mg/kg·d)能够促进免疫抑制小鼠的DTH反应,改变血清溶菌酶含量、提高脾脏和胸腺指数,并呈作用剂量依赖关系。在体外细胞实验中,LMWHA(25-200ug/mL)加强了小鼠腹腔巨噬细胞的吞噬活性以及一氧化氮合酶(iNOS)活性,提高了一氧化氮(NO)的生成量,促进了脾脏T淋巴细胞的增殖。由此可见LMWHA能够调节小鼠的细胞免疫和非特异性免疫功能。 Low molecular weight hyaluronan (LMWHA)were prepared from hyaluronan by using free radical degradation,and the immunostimulatory activities of LMWHA on macrophages and T lymphocytes were evaluated by using in vitro cell models and in vivo animal experiments.The effect of LMWHA on the delayed type hypersensitivity response (DTH)induced by DNFB, serum hemolysin activity, thymus and spleen indexes from immunosuppressed mice were detected.The LMWHA(20-]20 mg/kg, d)could induce DTH and increase the leveS of serum lysozyme and the indices of thymus and spleen in a dose - dependent manner. Furthermore, LMWHA (25-200μg/mL)could promote the proliferation of splenocytes,strengthen the phagocytic activity and induce the increase of nitric oxide (NO) production and the activity of nitric oxide synthase (iNQS) in mouse peritoneal macrophages.The results showed that LMWHA could regulate the immune function of mice.
