WNT信号通路抑制因子-1和β连环蛋白在肾透明细胞癌中的表达及临床意义 被引量:6

The expression and clinical significance of WIF-1 and β-catenin in clear-cell renal cell carcinoma

摘要 目的探讨WNT信号通路抑制因子-1(WIF-1)和β连环蛋白(β-catenin)在肾透明细胞癌(ccRCC)中的表达及其与肾癌临床分期、病理分级的相关性。方法选取2010年2月—2015年6月中国医科大学附属盛京医院第二泌尿外科行根治性手术切除的ccRCC患者102例为研究对象,应用免疫组织化学SP法检测102例肾细胞癌组织及相对应的癌旁正常肾脏组织中WIF-1、β-catenin表达情况。结果 WIF-1在ccRCC组织及正常组织中的表达差异有统计学意义(59.7±19.6 vs.112.2±18.6,t=3.104,P<0.05);β-catenin在ccRCC组织及正常组织中的表达差异亦有统计学意义(157.1±17.2 vs.102.1±18.7,t=4.015,P<0.05)。WIF-1的表达与肾透明细胞癌的临床分期(r=-0.542,P<0.05)、病理分级(r=-0.476,P<0.05)呈负相关;β-catenin的表达与肾透明细胞癌的临床分期(r=0.511,P<0.05)、病理分级(r=0.562,P<0.05)呈正相关.;WIF-1与β-catenin在肾癌中的表达呈负相关(r=-0.425,P<0.01)。结论 WIF-1表达降低和β-catenin表达升高与肾癌的病理分级和临床分期有关,对ccRCC预后评价有一定参考意义,也可为其治疗提供一种可能的靶向治疗途径。 Objective To investigate the expression of WTF-1(WNT inhibitory factor-1,WlF-1) and β-catenin in clear-cell renal cell carcinoma(ccRCC),and their correlations with clinical stage and pathological grading of renal cell carcinoma of renal cell carcinoma.Methods 102 patients with ccRCC who received radical resection in the Department of Urology,Shengjing Hospital of China Medical University from February 2010 to June 2015 were selected as the research subjects.Immunohistochemistry SP was used to detect the expression of WIF-1 and β-catenin in 102 cases of renal cell carcinoma tissues and the corresponding adjacent normal renal tissues.Results The difference of WIF-1 expression between ccRCC tissues and normal tissues were significandy statistical(5.97 ± 1.96 vs.112.2 ± 18.6,t =3.104,P 0.05),and β-catenin was(157.1 ±17.2 vs.102.1 ± 18.7,t=4.015,P 0.05).The WIF-1 expression was negatively correlated with the clinical stage(r =-0.542,P 0.05) and the pathological classification(r =-0.476,P 0.05) of ccRCC.The β-catenin expression was positively correlated with the clinical stage(r =0.511,P 0.05) and the padiological classification(r =0.562,P 0.05) of ccRCC.The expression of WIF-1 and B-catenin was negatively correlated in renal cell carcinomas(r =- 0.425,P 0.01).Conclusions The decreasing of WIF-1 expression and the increasing of β-catenin expression are related with the clinical stage and the padiological grading of RCC,and has a certain reference significance to the prognosis evaluation of ccRCC,and can provide a possible target therapy for the treatment of RCC.
