青蒿琥酯对手足口病单个核细胞分泌血管内皮生长因子的作用

摘要 目的探讨不同浓度青蒿琥酯对手足口病患者单个核细胞分泌VEGF的影响。方法采用Ficoll密度梯度离心法分离单个核细胞,收集单个核细胞进行培养,并加入不同浓度的青蒿琥酯,收集上清液,应用ELISA双抗体夹心法检测上清中VEGF含量,采用SPSS 19.0软件进行统计分析。结果与正常对照组比较,不同浓度青蒿琥酯作用下,细胞培养上清VEGF含量差异均有统计学意义(P<0.05),其中重症组VEGF浓度最高。普通组和重症组的VEGF浓度随着青蒿琥酯浓度的增加而降低,差异有统计学意义(P<0.05)。结论手足口病患儿VEGF水平偏高,重症患儿VEGF水平比普通手足口病患儿高。青蒿琥酯能下调手足口病患儿单个核细胞分泌VEGF。 Objective To explore the effect of artesunate in mononuclear cells secreting vascular endothelial growth factor (VEGF) in hand - foot - mouth disease(HFMD). Methods Ficoll - density gradient centrifugation was used to isolate mono- nuclear cells, and mononuclear cells were collected for culture ; adding different concentrations of artesunate, supernatants were collected ; content of VEGF in the supernatants was detected by ELISA double antibody sandwich method and was conducted by SPSS 19.0 software for statistical analysis. Results Compared to the normal control, the VEGF concentration difference had statistical significance(P 〈0.05) with the effect ofartesunate. The severe HFMD group was with the highest VEGF concentra- tion. In severe HFMD group, the VEGF concentration decreased with the increase of artesunate, and the difference was statisti- cally significant ( P 〈 0.05 ). Conclusion The VEGF concentration levels of HFMD children were relatively high ; that of the severe HFMD children were higher than that of the common HFMD children. Artesunate could down - regulate mononuclear cells secreting VEGF in HFMD children.
