摘要 目的通过分析盐酸乙醇的构成、理化性质,结合实验结果,研究其作用机制及原理,并提出在某些应用中的改良可能,帮助病理技术工作者更好应用。方法分析对比盐酸和乙醇的理化性质及相互作用,并设计实验验证其对苏木素染色的影响。结果盐酸乙醇分化液中乙醇的作用:1并非脱水;2使溶液p H值略微升高;3乙醇是一种很好的溶剂,能提高反应速率,但对苏木素的褪色作用不明显;4乙醇张力小,其内部溶质较活跃,扩散快;5增大了组织脱片的可能性。结论在常规苏木素-伊红(HE)染色中,盐酸乙醇分化液能够很好地完成分化作用,其中乙醇起到提高反应速率,加快溶质运动的作用。对于容易脱片的组织,可采用盐酸蒸馏水溶液。只要充分掌握盐酸乙醇的理化性质,就能更好地加以合理应用。 Objective To study the mechanism and principle of hydrochloric acid alcohol through the analysis of composition and physicochemical properties and experimental results, proposed the improvement in some applications, which may help the technician of pathology to do better. Methods Compared the physicochemical properties and interaction of hydrochloric acid and alcohol, and designed experiment to verify its effect on hematoxylin staining. Results The role of alcohol in differentiation of hydrochloric acid alcoholic: ① not for dehydrating; ② make the pH of the solution was slightly increased; ③ alcohol is a good solvent, can improve the reaction rate, but the fade effect on hematoxylin is not obvious; ④ the tension of alcohol is small, the solute in alcohol is active,and diffuse fast; ⑤increases the probability of tissue shedding. Conclusion In the routine HE staining, differentiation of hydrochloric acid alcohol solution can act well, the alcohol can enhance the reaction rate, speed up the movement of solution.,it can use aqueous hydrochloric acid for the tissue easy drop off. As long as a full grasp of the physicochemical properties of hydrochloric acid alcohol, it can be better applicated reasonably.
