The Genetic Diversity Analysis of Ponkan Using SSR and InDel Molecular Marker

摘要 利用SSR和InDel分子标记对来自不同国家与地区的26份椪柑资源进行了遗传多样性分析。21对引物共获得62个标记位点,产生了105条多态性条带。多态性标记位点的平均PIC值为0.32,平均期望杂合度为0.30。群体遗传结构和主坐标分析均表明,椪柑资源内部遗传多样性水平低,遗传变异较小。印度酸桔与椪柑遗传关系较远;Emperor、濑户见及早香等椪柑杂交后代,与椪柑存在明显遗传差异。采用筛选的分子标记组合(SSR14、SSR16、InDel2、InDel6、SSR5和SSR20)可有效鉴别椪柑材料。 SSR and InDel molecular markers were used to analyze genetic diversity of 26 ponkan germplasm resources.Twenty one pairs of primers produced 62 marker loci and 105 polymorphic PCR products with average 5amplified bands per pair of primers.The mean Shannon index of primers was 0.32 and average expected heterozygosity was 0.30.The results of population genetic structure and principal components analysis showed a low genetic diversity among different ponkan germplasm.Cleopatra sunki had far genetic relationship with ponkan.The hybrids of ponkan,such as Emperor,Setomi and Hayaka mandarin showed a significant genetic difference from ponkan.Selected molecular marker combinations,such as SSR14,SSR16,InDel2,InDel16,SSR5 and SSR20could be used to distinguish ponkan germplasm resources.
