摘要
目的探讨氧化苦参碱(OM)诱导人胃癌细胞SGC-7901的细胞凋亡及其机制。方法应用四甲基偶氮唑盐(MTT)法、荧光显微镜及流式细胞仪(FCM)检测不同浓度的OM在不同的时间对SGC-7901细胞的抑制,免疫组织化学法检测胃癌细胞中Bcl-2蛋白表达。结果 MTT法显示,随着OM浓度的梯增及时间的延长,细胞的抑制率亦逐渐增强,呈时间、剂量依赖性。荧光显微镜下可见到细胞调亡形态。流失细胞仪分析,OM可阻滞SGC-7901细胞在G0/G1期。免疫组化结果显示,不同药物浓度可明显阻滞Bcl-2蛋白的表达,Bcl-2蛋白的表达下调。结论氧化苦参碱(OM)对人胃癌细胞SGC-7901有显著的抑制作用,其机制可能与Bcl-2蛋白表达下调有关。
Objective To study the apoptosis of SGC-7901 gastric cancer cells induced by oxymatrine(OM)and its mechanism.Methods MTT assay,fluorescence microscopy and flow cytometry were used to detect the inhibition effects of OM on SGC-7901 cells at different concentrations and different time.The cellular expression of Bcl-2protein was detected by immunohistochemistry.Results MTT showed that the oxymatrine inhibited the growth of SGC-7901 and induced G0/G1 cell cycle arrest.Fluorescence microscopy showed that oxymatrine induced cell apoptosis.Bcl-2expression was downregulated.Conclusion Oxymatrine can induce apoptosis of gastric cancer cells SGC-7901.The mechanism is to reduce the expression of Bcl-2.
